Based on the Theory of Brain-gut Axis,to Explore the Effects of Guiling Pa'an Pill on Striatal DA and Serum CCK and VIP Levels in PD Rats
Objective Based on the brain-gut axis theory,the effects of Guilingpa'an Pill on the levels of dopamine(Dopa-mine,DA)in striatum lysate and serum cholecystokinin(cholecystokinin,CCK)and vasoactive intestinal peptide(vasoactive intestinal peptide,VIP)in PD rats were studied.Methods The PD rat model was made by injection of 6-hydroxydopamine(6-OHDA).The successful PD rats were randomly divided into model group(n=15),western medicine group(n=15),high dose group(n=15),middle dose group(n=15)and low dose group(n=15).The model group and sham operation group were given the same volume of normal saline,and the other groups were given corresponding drugs for 4weeks.Blood samples were taken from abdominal aorta of rats,and the levels of DA in striatum lysate and serum CCK and VIP were detected by enzyme-linked im-munosorbent assay(ELISA).Results Compared with sham operation group,the levels of striatum DA,serum CCK and VIP in model group,western medicine group and each dose group were significantly lower than those in sham operation group(P<0.01);Compared with the model group,the levels of striatum DA,serum CCK and VIP in the western medicine group and each dose group were significantly higher than those in the model group(P<0.01).Conclusion Guiling Pa'an Pill has obvious neuro-protective effect,and its mechanism is related to activating brain-gut axis,increasing the level of CCK and VIP,increasing the level of DA in substantia nigra and striatum of midbrain,and improving intestinal function.
