Effect of acupuncture pretreatment on gastric mucosal status and Gli 1/Gli 2/Sufu signaling pathway in rats with gastric ulcer
Objective:To investigate the effects of acupuncture pretreatment on gastric mucosal status and glioma-associated oncogene homolog 1(Gli1)/glioma-associated oncogene homolog 2(Gli2)/serine/serine signaling pathway in gastric ulcer rats.To investigate the effect of acupuncture on the prevention of gastric mucosal damage and to provide experimental basis and theoretical support for acupuncture in the clinical treatment of gastric ulcer disease,we investigated the effects of the Fused inhibi-tor of threonine kinase(Sufu)signaling pathway.Methods:52rats were randomly divided into normal group,model group,mox-ibustion pretreatment group,the acupuncture preconditioning group,each group of 13.The rats in the normal and model groups were fixed without treatment;the rats in the moxibustion pretreatment group were subjected to moxibustion at the Zusanli point and the Zhongwan point for 20min per day,moxibustion once a day for 7days.After pretreatment,rats in the model group,moxibus-tion pretreatment group and acupuncture pretreatment group were gavaged with anhydrous ethanol combined with aspirin suspen-sion to establish a model of acute gastric mucosal lesion.The general condition of rats and histopathological changes of gastric mu-cosal injury were observed;the concentrations of superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxi-dase(GPX)in rat serum were measured by enzyme-linked immunosorbent assay(ELISA),and the expression of Gli 1,Gli 2and Sufu proteins in rat gastric mucosa was analyzed by protein blotting(Western blot).Results:In the model group,incom-plete structural destruction of epithelial cells and significant damage to gastric mucosal tissues were seen,and the UI index score was significantly higher than that of the normal group(P<0.05).Compared with the normal group,serum MDA and GPX con-centrations were increased(P<0.05)and SOD concentrations were decreased(P<0.05)in the model group;compared with the model group,MDA and GPX concentrations were decreased(P<0.05)and SOD concentrations were increased(P<0.05)in the acupuncture pretreatment group;compared with the normal group,Gli 1,Gli2and Sufu protein expressions were significantly in-creased(P<0.05)in the model group;compared with the model group,Gli 1,Gli2and Sufu protein expressions were significantly decreased(P<0.05)in the acupuncture pretreatment group.Compared with the normal group,Gli1,Gli2and Sufu protein ex-pressions were significantly increased in the model group(P<0.05);compared with the model group,Gli1,Gli2and Sufu protein expressions were significantly decreased in the acupuncture pretreatment group(P<0.05).Conclusion:Acupuncture pretreat-ment can change the gastric mucosal status of gastric ulcer rats,and the mechanism may be related to the activation of Gli 1/Gli 2/Sufu signaling pathway.
