The Study of Chaihu Jialonggu Oyster soup on Improving Insomnia and Associated Bad Mood in Sleep Deprived Rats
Objective:To observe the curative effect of Chaihu Jialonggu Oyster soup on insomnia and accompanying bad mood in sleep deprived rats,and its effect on 5-hydroxytryptamine(5-HT)and norepinephrine(NE),and explore its mecha-nism.Methods:70healthy SD male rats were randomly divided into blank group,model group,diazepam group,escitalopram group,and low,medium and high dose groups of Chaihu plus Longgu Oyster soup according to body weight,with 10rats in each group.The blank group did not undergo sleep deprivation,and the other six groups were subjected to sleep deprivation for a period of 21 days using an improved multi platform water environment method.After successful modeling,the blank group and model group were gavaged with equal amounts of physiological saline for 14days.The diazepam group,escitalopram group,and low,me-dium,and high dose groups of traditional Chinese medicine were gavaged according to the dosage given.Observation indicators in-clude changes in the general condition of rats,detection of sleep latency and duration using pentobarbital sodium sleep synergy ex-periment,open field experiment,sucrose water preference experiment to detect behavioral differences in rats,and ELISA to de-tect the levels of 5-HT and NE in rat serum.Result:The hair color of sleep deprived rats is dim,and their alertness,responsive-ness,irritability,and aggressiveness to environmental stimuli are enhanced.The sleep latency of sleep deprivation rats was signifi-cantly longer than that of the blank group(P<0.05),and the sleep duration was significantly shorter than that of the blank group(P<0.05),indicating that the sleep deprivation model was successfully established.After intervention,the sleep latency time of the low,medium,and high dose groups of traditional Chinese medicine was significantly shorter than that of the model group(P<0.05),while the sleep duration of the medium and high dose groups of traditional Chinese medicine was longer than that of the model group(P<0.05).The scores of open field experiments in the middle and high dose groups of traditional Chinese medicine were significantly higher than those in the model group(P<0.05).The preference rate for sucrose water in the low,medium,and high dose groups of traditional Chinese medicine was higher than that in the model group,but there was no statistical difference.The serum levels of 5-HT and NE in the middle dose group of traditional Chinese medicine were significantly higher than those in the model group(P<0.05).Conclusion:Chaihu Jialonggu Oyster soup can shorten the sleep latency and prolong the sleep dura-tion of sleep deprivation model rats,and has the effect of improving insomnia in rats.It can improve the scores of open field exper-iments and the preference rate for sucrose water in rats,and has an improvement effect on the lack of pleasure and negative emo-tions in rats.It can upregulate the levels of 5-HT and NE in the serum of sleep deprived rats.The above shows that Chaihu Jia-longgu Oyster soup has a significant effect on improving insomnia and accompanying bad mood in sleep deprived rats,and its mechanism may be related to the increase of serum 5-HT and NE content.
