Effect ofSanbaohutong Decoction on the immune imbalance of Th17/Treg cells in young rats with allergic rhinitis
Objective:To study the effect of Sanbaohutong Decoction on the immune imbalance of Th17/Treg cells in young rats with allergic rhinitis.Methods:Thirty-two SD female rat pups were randomly divided into blank group,model group,Sanbao-hutong Decoction group,and desloratadine group,with 8rats in each group.Except for the blank group,all the other groups used smoke+Dahuang bitter cold diarrhea method to establish the lung spleen deficiency syndrome model,and used intraperitoneal in-jection of ovalbumin(OVA)and aluminium hydroxide Al(OH)3method for basic sensitization,and then used OVA nasal drops method for local sensitization to establish the allergic rhinitis model.After successful modelling,the blank group,the model group,Sanbaohutong Decoction and the desloratadine group were gavaged with saline,saline,Sanbaohutong Decoction and deslo-ratadine dry suspension for 14days,respectively.We observed the basic behavioural changes of the young rats,scored the nasal symptoms,observed the histopathological changes of the nasal mucosa by HE staining,and detected the levels of interleukin-17and transforming growth factor β(TGF-β)in serum by ELISA,and the protein expression level of RORγt in spleen by West-em blot.Results:Compared with the blank group,the nasal mucosa of the model group had damaged epithelial structure,disor-ganized cilia,epithelial edema,and infiltration of cup cells and eosinophils;compared with the model group,the nasal symptom scores of the Sanbaohutong Decoction group and the desloratadine group were reduced after administration of Sambucus jelly(P<0.05),and the epithelial cilia of the nasal mucosa tissues were more neat,the epithelial edema was reduced,and the infiltration of cup cells and eosinophils was decreased;compared with the model group,the nasal mucosa of the Sanbaohutong Decoction group and desloratadine group had reduced epithelial cilia,and the infiltration of cup cells and eosinophils.Compared with the model group,the IL-171evel and the expression of RORγt protein in spleen were reduced in Sanbaohutong Decoction and deslora-tadine group(P<0.05),and the level of TGF-β was increased(P<0.05).Conclusion:Sanbaohutong Decoction can improve nasal symptoms and reduce inflammatory cell infiltration in young rats with allergic rhinitis with lung spleen deficiency syndrome,and its mechanism may be related to the regulation of the immune imbalance of Th17/Treg cells and the expression of related pro-teins.
