The regulatory effect of Chuanxiong-Duhuo-Achyranthes in the immune balance of TH17 Treg cells in the patho-genesis of osteoarthritis
Objective:To investigate the regulatory effect of the combination of Chuanxiong-Duhuo-Achyranthes on the im-mune balance of Th17/Treg cells in the pathogenesis of osteoarthritis.Methods:40SPF grade sterile C57BL/6mice were pur-chased,and 10mice were randomly selected as blank control group;the remaining 30mice were constructed by Hulth-Telhag method to establish an animal model of knee arthritis,and randomly divided into model group,Isovaleric acid group and Chuanx-iong-Duhuo-Achyranthes compatibility group,each with 10animals.The blank control group and the model group were given in-traperitoneal perfusion with equal volume of normal saline,the isovaleric acid group was given isovaleric acid by intraperitoneal gavage,and the Chuanxiong-Duhuo-Achyranthes compatibility group was given the Chuanxiong-Duhuo-Achyranthes decoction by intraperitoneal administration.Each group lasted for 7days.After intervention,HE staining results,inflammatory factor levels and Th17/Treg cell immunity were compared in each group.Results:Under HE staining,the isovaleric acid group was accompa-nied by mild articular cartilage destruction,and a little inflammatory cell infiltration was seen;the articular cartilage destruction was not obvious in the Chuanxiong-Duhuo-Achyranthes combination group,and there were relatively few inflammatory cells;Chuanxiong-Duhuo-Achyranthes IL-17,IL-6and IL-4in the compatibility group were lower than those in the isovaleric acid group(P<0.05);IL-10and TGF were higher than those in the isovaleric acid group(P<0.05);the results of flow cytometry showed that:After 7days of intervention,the Th17cells in the Chuanxiong-Duhuo-Achyranthes compatibility group were lower than those in the isovaleric acid group and the model group(P<0.05);the Treg cells were higher than those in the isovaleric acid group and the model group(P<0.05).Conclusion:The combination of Chuanxiong-Duhuo-Achyranthes for osteoarthritis can re-duce the knee joint synovium and soft tissue damage,and reduce the level of inflammatory factors in the body.The mechanism may be related to the regulation of Th17/Treg cell immune balance.
