Study on the Inflammatory Response and Therapeutic Effect of Qingyingtang on LCWE-induced Kawasaki Disease Model Mouse Based on TLR4and NF-κB Signaling Pathway
Objective:To study the therapeutic effect of Qingyingtang(QYT)on Kawasaki disease(KD)model mouse.Methods:50eligible C57BL/6JSD mice were randomly divided into blank control group(NC),Kawasaki disease model group(KD),QYT low-dose group(QYT-L),QYT high-dose group(QYT-H),and aspirin group(ASC),with 10mice in each group.mouse except for the NC group were modeled by lactobacillus casei cell wall extract(LCWE).After successful modeling,50mg/kg QYT,100mg/kg QYT and 50mg/kg ASC were given by gavage in the QYT-L,QYT-H and ASC groups,respectively,and an equal volume of PBS was given by gavage in the NC group and the KD group,1t/d,for 7d.Changes in body weight of the mice were recorded;cardiac ultrasonography was used to examine the LVEF and FS;enzyme-linked immunosorbent assay(ELISA)to determine the serum levels of IL-6,IL-1β,and TNF-α in mice;hemagglutination analyzer and ELISA to detect plasma fibrinogen(FIB),D-dimer,and antithrombin Ⅲ(AT-Ⅲ);observation of hematoxylin-eosin(HE)staining of coro-nary arteries of mouse heart tissue for pathomorphic changes and The extent of lesions was determined by IPP;protein western-blotting(WB)was used to detect the expression of NF-Κb p65,p-NF-Κb p65,TNF-α,TLR4,and MyD88proteins in the cor-onary tissues of mouse hearts.Results:After treatment,the body weight,LVEF,FS,and plasma FIB levels of mouse in the QYT-L,QYT-H,and ASC groups were significantly higher than those in the KD group(P<0.05);serum TNF-α,IL-6,and IL-1β levels,D-Dimer,and AT-Ⅲ levels were significantly lower than those in the KD group after intervention in the QYT-L,QYT-H,and ASC groups(P<0.05);and KD group Coronary artery wall thickening with inflammatory infiltration was slightly improved in the QYT-L group and significantly improved in the QYT-H and ASC groups,and the results of IPP measurement of lesion area showed that the QYT-L,QYT-H,and ASC groups were significantly lower than that of the KD group(P<0.05);compared with the NC group,the expression of p-NF-Κb p65,TNF-α,TLR4,and MyD88proteins was significantly increased in the KD group(P<0.05);compared with the KD group,p-NF-Κb p65,TNF-α,TLR4,and MyD88protein expression in mouse in the QYT-L,QYT-H,and ASC groups decreased significantly(P<0.05).Conclusion:Qingyingtang can effectively improve myocardial function and coagulation function,inhibit the activation of TLR4/NF-Κb signaling pathway and thus inhibit the release of inflammatory factors,thus exerting therapeutic effects on coronary artery injury in KD mice.
