Objective:To explore the effect of Modified Zuo-gui-wan on the bone microstructure of ovariectomized osteoporo-sis model mice and its possible mechanism of action,in order to provide a basis for the treatment of osteoporosis with the method of Bu-shen-yi-qi-huo-xue.Method:218-week-old C57BL/6J female mice were randomly divided into a model group,a sham surgery group,and Modified Zuo-gui-wan group,with 7mice in each group.The model group and Modified Zuo-gui-wan group mice established osteoporosis models by removing both ovaries,while the sham surgery group mice removed an equal amount of adipose tissue.The Modified Zuo-gui-wan group was gavaged with Modified Zuo-gui-wan solution,while the model group and sham surgery group were gavaged with an equal amount of distilled water.After 8weeks of administration,bone tissue images were collected using mouse Micro CT combined with HE staining tissue sectioning technology to quantitatively analyze the trabecular bone and evaluate the effect of Modified Zuo-gui-wan on bone microstructure.Result:Observation of microstructure of mouse fe-mur:The overall differences in bone volume fraction(BV/TV,%),trabecular thickness(Tb.Th,mm),and trabecular separa-tion(Tb.Sp,mm)among the three groups of mice were statistically significant(P<0.05);The difference in bone volume frac-tion,trabecular thickness,and trabecular separation between the sham surgery group and the model group was statistically signifi-cant(P<0.05);Compared with the model group,the difference in bone volume fraction and trabecular thickness between the Modified Zuo-gui-wan group and the model group was statistically significant(P<0.05),while there was no statistically signifi-cant difference in trabecular separation(P>0.05);There was no statistically significant difference in bone volume fraction,tra-becular thickness,and trabecular separation between the Modified Zuo-gui-wan group and the sham surgery group(P>0.05).Observation results of femoral tissue morphology in mice:Overall comparison of the percentage of bone trabecular area(%)a-mong the three groups of mice showed statistically significant differences(P<0.05).The differences between the sham surgery group and the model group,as well as the Modified Zuo-gui-wan group and the model group,were statistically significant(P<0.05).However,there was no statistically significant difference between the sham surgery group and the Modified Zuo-gui-wan group(P>0.05).Conclusion:the Modified Zuo-gui-wan can partially reverse bone loss and bone microstructure damage in OVX induced osteoporosis mice,providing experimental basis for the treatment of osteoporosis with the method of supplementing Bu-shen-yi-qi-huo-xue.
