慢性萎缩性胃炎及其中医寒、热证尿液代谢组学研究

目的:通过尿液代谢组学分析慢性萎缩性胃炎患者与健康人群的差异代谢物及代谢通路,继而阐述尿液代谢组学未能佐证慢性萎缩性胃炎中医寒、热辨证存在差异物质基础的原因.本研究收集慢性萎缩性胃炎患者以及健康志愿者的尿液样本,使用核磁共振氢谱技术定量检测其代谢物的含量,运用PCA、PLS-DA以及OPLS-DA进行多元统计分析.结果显示:1.慢性萎缩性胃炎患者与健康对照组相比较:尿液中黄嘌呤、马尿酸、柠檬酸含量升高,肌酐、组胺酸、肌酸、鸟氨酸、磷酸肌酸的含量降低,差异具有统计学意义(P<0.05);2.慢性萎缩性胃炎寒、热证尿液中未找到具有明显差异的代谢物.结论:1.与健康人群相比,慢性萎缩性胃炎患者尿液中差异代谢物主要与氨基酸代谢、三羧酸循环、尿素循环等代谢途径相关;2.慢性萎缩性胃炎寒、热证之间未找到明显差异的代谢物,原因可能与慢性萎缩性胃炎疾病与证候的特点、尿液样本的选取和处理、辨证诊断和实验过程中的误差相关.
Study on urine metabolomics of chronic atrophic gastritis and its TCM cold and heat syndrome
Objective:The aim of this study is to analyze the differential metabolites and metabolic pathways in the urine of patients with chronic atrophic gastritis(CAG)compared to a healthy population through Urinary Metabolomics.Subsequently,we elucidate the reasons why Urinary Metabolomics cannot substantiate the material basis for the traditional Chinese medicine differ-entiation of cold and heat syndromes in chronic atrophic gastritis.Methods:Urine samples were collected from chronic atrophic gastritis and healthy volunteers.Quantitative analysis of urine metabolites was conducted using proton nuclear magnetic resonance(1H-NMR).Multivariate statistical analyses,including Principal Component Analysis(PCA),Partial Least Squares Discrimi-nant Analysis(PLS-DA),and Orthogonal Partial Least Squares Discriminant Analysis(OPLS-DA),were employed.Results:1.Compared with healthy people,the metabolites in urine of CAG patients were mainly related to amino acid metabolism,tricar-boxylic acid cycle and urea cycle.2.The reason why there is no obvious difference in urine metabolites between CAG spleen-stomachdeficiency cold and spleen-stomachdampness-heat syndrome may be related to the characteristics of CAG disease and syndrome,the selection and treatment of urine samples,the dialectical diagnosis and the errors in the experimental process.
