Tumor suppressive effects of Huangqi injection in thyroid cancer-bearing mice and its impact on the expression of tis-sue apoptosis related proteins and NF-κB pathway
Objective:To analyze the tumor suppressive effects of Huangqi injection in thyroid cancer-bearing mice,and its im-pact on the expression of tissue apoptosis related proteins and nuclear factors-κB(NF-κB)pathway.Methods:50mice were ran-domly divided into the model control group,the(low-dose,medium-dose and high-dose)Huangqi injection groups,and the cispl-atin group,with 10mice in each group.For each group,thyroid cancer-bearing mouse models were established and in vivo anti-tumor experiments were conducted.Mice in the model control group were given gavage with 20ml/kg of normal saline every day.Mice in the(low-dose,medium-dose and high-dose)Huangqi injection groups were respectively given intraperitoneal injection of 4.0g/kg,8.0g/kg and 12.0g/kg of Huangqi injection,for 20d.Mice in the cisplatin group were given intraperitoneal injection of cisplatin(2.5× 10-3 g/kg))on day 1 and day 11,and were given gavage with 20ml/kg of normal saline at other time.Tumor volume was re-corded.20d later,the mice were sacrificed and tumor tissues were collected to calculate the tumor inhibition rate.The protein expres-sion of B cell lymphoma-2-associated X protein(Bax),B cell lymphoma-2(Bcl-2),caspase-3and NF-κBp65was detected by immunohistochemical method.Results:Compared with the model control group,tumor weight and volume of the(low-dose,medi-um-dose and high-dose)Huangqi injection groups were significantly lower(P<0.05),showing certain dose-dependent effect.The tumor inhibition rates in the(low-dose,medium-dose and high-dose)Huangqi injection groups were 9.62%,22.75%and 44.87%,respectively.Compared with the model control group,the protein expression of Bax and caspase-3in tumor tissues of other groups was significantly up-regulated,while the expression of Bcl-2protein was significantly down-regulated(P<0.05).The pro-tein levels of Bax and caspase-3in the(low-dose,medium-dose and high-dose)Huangqi injection groups increased,while the level of Bcl-2protein decreased with the increase of dose.The differences between any groups were statistically significant(P<0.05).Compared with the model control group,the expression of NF-κBp65protein in tumor tissues of other groups was significantly down-regulated(P<0.05).Compared with cisplatin group,the expression of NF-κBp65 protein in tumor tissue of Huangqi injection(low,medium and high dose)group was up-regulated(P<0.05).The protein of NF-κBp65in Huangqi injection(low,medium and high dose)group decreased with the increase of concentration(P<0.05).Conclusion:Huangqi injection can inhibit the prolif-eration of thyroid cancer.The mechanism may be regulating the expression of apoptosis related proteins and NF-κB signaling path-way.
