榆苋方对大肠湿热型溃疡性结肠炎大鼠肠道微生态及血清VIP、CCK、IL-17、IL-23、IL-10表达的影响

目的:观察榆苋方对大肠湿热型溃疡性结肠炎大鼠肠道微生态及血清VIP、CCK、IL-17、IL-23、IL-10 表达的影响,探讨其治疗溃疡性结肠炎可能的作用机制.方法:将 48 只健康SD大鼠随机分为 6 组:空白组、模型组、低剂量组、中剂量组、高剂量组及柳氮磺嘧啶组,每组各 8 只.空白组不造模,造模组通过三硝基苯磺酸(TNBS)法制备溃疡性结肠炎大鼠模型.造模成功后,空白组、模型组给予以生理盐水直肠内给药;小剂量治疗组、中剂量治疗组、高剂量治疗组分别使用榆苋方不同剂量药物保留灌肠,柳氮磺嘧啶组予柳氮磺吡啶灌肠,均连续直肠内给药 14d.采用菌群培养观察各组肠道微生态情况(双歧杆菌、乳酸杆菌、肠球菌、肠杆菌),气相色谱法检测各组大鼠结肠肠道中乙酸、丙酸、丁酸水平,ELISA法测定大鼠血清VIP、CCK、IL-17、IL-23、IL-10 含量的变化.结果:与空白对照组比较,模型组、榆苋方组及柳氮磺嘧啶组双歧杆菌、乳酸杆菌、乙酸、丙酸、VIP、CCK、IL-10 均降低,IL-17、IL-23 表达升高,经榆苋方治疗后,随着给药浓度的增加,双歧杆菌、乳酸杆菌、乙酸、丙酸、VIP、CCK、IL-10 均明显升高(P<0.05),IL-17、IL-23 表达均明显降低(P<0.05).结论:榆苋方对大肠湿热型溃疡性结肠炎大鼠的肠道微生态、短链脂肪酸及脑肠肽有较好的调节作用.
Effect of Yuxian Decoction on intestinal microecology and serum VIP,CCK,IL-17,IL-23and IL-10expression in rats with ulcerative colitis of damp-heat type in large intestine
Objective:To investigate the effects of Yuxian Decoction on the intestinal microecology and the expression of VIP,CCK,IL-17,IL-23,IL-10in serum of rats with large intestine damp heat type ulcerative colitis to explore its possible mechanism of action of colitis.Methods:Forty-eight healthy Sprague-Dawley rats were randomly divided into six groups(blank control group,mod-el group,low dose treatment group,medium dose treatment group,high dose treatment group and Salazosulfadimidine group,eight rats in each group).The blank control group did not model,the rats in model group were used to prepare a rat model of ulcerative coli-tis by trinitro benzenesulfonic acid(TNBS).After successful modeling,the blank control group,the model group were given an equal dose of physiological saline,and each administration group was given low dose,medium dose and high dose Yuxian decoction and the sulfasalazine suspension rectally continuous rectally for 14d.The intestinal microecology in each group was observed by microflora cul-ture,including Bifidobacterium,Lactobacillus,Enterococcus and Enterobacter.The levels of acetic acid,propionic acid and butyric acid in intestinal tract of colon of mice in each group were detected by gas chromatography method.The contents of serum brain-gut peptide such as VIP,CCK,IL-17,IL-23,IL-10were detected by ELISA.Results:Compared with the blank control group,the contents of Bifidobacterium,Lactobacillus,acetic acid,propionic acid,VIP,CCK,IL-10were reduced in the model group,SASP group and each dose groups of Yuxian Decoction,and the level of IL-17,IL-23were increased,after treating with Yuxian Decoction,with the gradual increase of the administered dose,the contents of Bifidobacterium,Lactobacillus,acetic acid,propionic acid,VIP,CCK,IL-10was significantly increased(P<0.05),and the IL-17,IL-23level was significantly reduced(P<0.05).Conclusion:Yuxian Decoction shows good regulation on the intestinal microecology and short-chain fatty acids and brain-gut peptide in large intes-tine damp heat type ulcerative colitis.
