Based on the identification and analysis of alternative splicing genes from RNA-Seq of the mammary gland tissue of Laoshan dairy goats at the early lactation,peak lactation and late lactation,a total of 151 503 alternative splicing events,corresponding to 17 627 alternative splicing genes,and 6 323 differentially splicing genes,significantly enriched to related GO entries including organelles,protein binding,mRNA processing and RNA splicing,and mainly involved in oxytocin signaling pathway,calcium signaling pathway,and protein processing in the endoplasmic reticulum,and KEGG pathway closely related to the physiological activity of breast tissue,and screened PTEN as the core gene involving in the regulation network of mammary gland development and lactation-related physiological processes.The results indicated that variable splicing has developmental stage specificity in mammary gland tissues of dairy goats at different lactation stages,the unique variable splicing gene is closely related to the physiological activity of mammary gland tissue and plays an important role in regulating the development of mammary gland tissue and lactation physiology of dairy goats.Our study can provide the theoretical basis for exploring the molecular mechanism of alternative splicing mechanisms regulating mammary gland development and lactation traits in dairy goats.
