摘要

为探究不同生活环境下锦鲤肠道微生物的差异,本研究采用16S rRNA高通量测序技术对野生和养殖锦鲤肠道微生物组成进行分析.结果表明,锦鲤肠道微生物中的优势门为变形菌门(Proteobacteria,29.81%)、蓝细菌门(Cyanobacteria,25.57%)、绿弯菌门(Chloroflexi,18.16%);不同环境下锦鲤肠道菌群在数量和群落结构上都有明显差异,在属水平上,养殖环境下优势菌属有红细菌属(Rhodobacter)、藤黄色杆菌属(Luteolibacter)、泉发菌属(Crenothrix),野生环境下优势菌属有绿丝菌属(Chloronema)、纤发鞘丝蓝细菌属(Leptolyngbya)、暖绳菌属(Caldilinea);在种水平上,养殖环境下优势菌有Methylobacterium adhaesivum、琥珀黄杆菌(Flavobacterium succinicans)、胭脂甲杆菌(Methylobacterium komagatae)、湖泊玫瑰单胞菌(Roseomonas lacus);野生环境下优势菌有空腔污水球菌(Defluviicoccus vanus)、Proteocatella sphenisci、湖泊玫瑰单胞菌、Roseomonas massiliensis、索氏鲸杆菌(Cetobacterium somerae);α多样性分析显示养殖环境下锦鲤肠道微生物的多样性以及物种丰富度均比野生样本高;肠道微生物功能预测显示,L1、L2、L3水平最富集的KEGG通路分别是新陈代谢、氨基酸代谢和萜类和类固醇的生物合成,不同环境下差异显著的KEGG通路包括2-氧羧酸添加代谢、甜菜碱生物合成、安莎霉素的生物合成、双酚降解、丁酸盐代谢等.本研究从肠道微生物角度探究了锦鲤的环境适应性,为其健康养殖提供了新思路.

Abstract

In order to investigate the differences of intestinal microbiota of Cyprinuscarpio haematopterus in different environments,16S rRNA high-throughput sequencing technology was employed to analyze the intestinal microbiota composition of wild and cultured C.haematopterus.The results revealed that Proteobacteria(29.81%),Cyanobacteria(25.57%),and Chloroflexi(18.16%)were the dominant phyla in intestinal microbiota of C.haematopterus.There were significant variations observed in both the abundance and community structure of intestinal flora across different environments.At the genus level,Rhodobacter,Luteolibacter,and Crenothrix were found to be dominant bacterial genera in cultured C.haematopterus,while Chloronema,Leptolyngbya,and Caldilinea were dominant bacterial genera in wild samples.Furthermore,Methylobacterium adhaesivum,Flavobacterium succinicans,Methylobacterium komagatae,and Roseomonas lacus were identified as dominant bacteria species within a culture environment;whereas Defluviicoccus vanus,Proteocatella sphenisci,Roseomonas lacus,Roseomonas massiliensis,and Cetobacterium somerae prevailed as dominant bacteria species within a wild environment.Alpha diversity analysis indicated that cultured samples exhibited higher biodiversity and species richness compared to their wild counterparts.Functional prediction of intestinal microbiota suggested that Metabolism,Amino Acid Metabolism,and Terpenoid and Steroid Biosynthesis were the most enriched KEGG pathways at L1,L2,and L3 levels,respectively.Significantly different KEGG pathways between different environments included 2-Oxocardboxylic Addmetabolism,BetalainBiosynthesis,Biosynthesisof Ansamycins,Bisphenol Degradation,and Butanoate Metabolism.This study explores the environmental adaptability of C.haematopterus from an intestinal microbiota perspective and provides novel insights for its healthy breeding.
