摘要

为了解黄精根状茎转录组中简单重复序列SSR的分布特点,充分开发SSR分子标记,本研究对黄精根状茎进行转录组测序,SSR位点分析与引物设计.以获得的61 116条Unigene为试材,采用MISA软件检索出黄精的SSR位点,利用Primer 3软件对检索到的SSR基序进行引物设计.结果表明:黄精转录组中检索出30 541个SSR位点,其发生频率分别为49.97%.单核苷酸和二核苷酸重复是黄精SSR的主要基序类型,以A/T和AG/CT基序的出现频率最高,占SSR总数的68.84%.通过引物设计,最终在黄精转录组中获得15 842对引物.因此,我们认为黄精根状茎转录组SSR位点的出现频率较高,为黄精的遗传多样性分析、分子育种等研究提供参考.

Abstract

To understand the distribution characteristics of simple repetitive sequences(SSRs)in the rhizome transcriptome of Polygonatum sibiricum tubers and to fully develop SSR molecular markers,this study conducted transcriptome sequencing,SSR site analysis,and primer design on Polygonatum sibiricum tubers.Using 61 116 unigenes obtained as test materials,we used MISA software to identify SSR loci in Polygonatum sibiricum,and the Primer 3 software was used to design primers for the identified SSR motifs.The results showed that 30 541 SSR loci were detected in the Polygonatum sibiricum transcriptome,with an occurrence frequency of 49.97%.Mononucleotide and dinucleotide repeats are the main motif types of Polygonatum sibiricum SSR,with A/T and AG/CT motifs having the highest frequency of occurrence,accounting for 68.84%of the total SSR.Through primer design,a total of 15 842 pairs of primers were ultimately obtained from the Polygonatum sibiricum transcriptome.Therefore,we believe that the occurrence frequency of SSR loci in the transcriptome of Polygonatum sibiricum is high,providing a reference for studies on genetic diversity analysis and molecular breeding of Polygonatum sibiricum.
