摘要
为克服Arnon公式法测定植物体系中叶绿素a和b以及β-胡萝卜素含量的干扰问题.论文基于可见光谱量测,引入多元曲线分辨-交替最小二乘(MCR-ALS)算法提取单分析物信号、使用朗伯-比尔定律计算浓度,建立了一种测定植物体系中叶绿素a和b以及β-胡萝卜素的定量分析方法,将发展的方法应用于烟草工业烘烤及烟叶醇化过程中3 种光合色素的抗干扰定量分析,并对弹簧烟杆装烟烘烤及烟夹装烟烘烤的实际体系进行了加标验证实验.研究结果表明,预测得到叶绿素a和b以及β-胡萝卜素的平均相对误差分别为1.9%和0.2%以及1.2%,0.8%和1.5%以及3.6%,实验结果满足实际检测的需求.论文提出的方法可为复杂植物体系中叶绿素a和b以及β-胡萝卜素含量的测定提供一种新的选择.
Abstract
In order to overcome the interference of Arnon formula method in the determination of chlorophyll a,chlorophyll b and β-carotene content in plant system.In this paper,based on visible spectrum measurement,the multivariate curve resolution-alternating least squares(MCR-AIS)algorithm was introduced to extract the single analyte signal from the mixed signal of the analysis system,and then the Lambert-Beer's law was used to calculate the concentration,an analysis method with some advantages of rapid,simple,low-cost,anti-interference,accurate quantitative was estimated for the determination of chlorophyll a,chlorophyll b and β-carotene in complex plant systems,and the proposed method was applied to the practical analysis system of tobacco industrial baking and tobacco aging process.In addition,the standard addition verification experiments were carried out on the actual system of flue-cured tobacco baking with spring tobacco rod loading method and tobacco clip loading method,the average relative errors of chlorophyll a,chlorophyll b and β-carotene were predicted to be 1.9%,0.2%and 1.2%,0.8%,1.5%and 3.6%,respectively.The experimental results meet the needs of actual detection.The method proposed in this paper could provide a new choice for the determination of chlorophyll a,chlorophyll b and β-carotene in complex plant system.
