植物组织培养褐化现象的发生及防治措施

褐化是影响植物组织培养最突出的问题,其严重影响培养物的生长和分化,甚至造成植物死亡.国内外褐化研究结果显示,褐化发生机制主要是酚类物质积累和氧化引起的氧化褐化,影响因素主要有植物品种和基因型、外植体的生理条件、培养环境、消毒方法及其他组培操作手段等,基于发生机制和影响因素提出的防治措施是选择不易发生褐化的外植体材料、优化培养基成分和培养条件、添加添加剂抑制或减轻褐化程度等.目前关于褐化发生机制的研究尚不能完全解释氧化褐化,褐化防治措施可以一定程度上控制褐化现象,但可能会对组培其他指标存在不利影响,实践中应根据实际情况筛选控制褐化的方法.
Occurrence and Control Measures of Browning in Plant Tissue Culture
Browning is the most prominent problem affecting plant tissue culture,which seriously affects the growth and differentiation of culture,and even causes plant death.The research results at home and abroad showed that the mechanism of browning was mainly oxidative browning caused by phenols accumulation and oxidation,and the main influencing factors were plant varieties and genotypes,physiological conditions of explants,culture environment,disinfection methods and other tissue culture methods.The prevention and control measures based on the occurrence mechanism and influencing factors of browning were to select explant materials,which were not easy to browning,optimize the medium compositions and culture conditions,and add additives to inhibit browning.At present,the research on the mechanism of browning could not fully explain the oxidative browning,and the control measures may have adverse effects on other indexes of tissue culture.
