Pathogen Identification of Dictyophora rubrovalvata Green Mold Disease and Screen of Green Fungicides
To identify the pathogenic species casuing green mold disease in Dictyophora rubrovalvata in Nayong County,Guizhou Province, and to assess the efficacy of fungicides in inhibiting the pathogen indoors while ensuring the safety of D.rubrovalvata mycelium, this study aims to establish a theoretical foundation for controling D.rubrovalvata green mold disease in the field. In this study,we isolated and purified the pathogenic fungus from the green mold in D.rubrovalvata exhibiting typical symptoms at the D.rubrovalvata planting base in Huangjia Village,Lezhi Town,Nayong County,Guizhou Province. The pathogenicity of the isolated pathogens was subsequently determined by Koch's method. The pathogenic strains were identified by morphological characterization, analysis of 18S rDNA and TEF1 gene sequences, and phylogenetic tree evolution.Eight commonly used fungicides were chosen to assess the virulence of the pathogen and evaluate safety of these fungicides on the mycelium of D.rubrovalvata.The study identified Trichoderma harzianum as the pathogenic fungus causing green mold disease in D.rubrovalvata.In indoor virulence assays, the most potent inhibitory effect on D.rubrovalvata growth was observed with 450 g/L prochloraz EW and 10%difenoconazole WG,exhibiting EC50 values of 0.0053 mg/L and 0.0007 mg/L,respectively.Evaluation of indoor safety for D.rubrovalvata, revealed that 80%zineb WP and 75%chlorothalonil WP exhibited the lowest inhibition rate on D.rubrovalvata mycelium, with EC50 values of 12.2854 mg/L and 10.8939 mg/L,respectively,followed by 25%procymidone In summary,25%procymidone Thiram WP exhibited potent inhinition against the D.rubrovalvata green mold disease, while also demonstrating a favorable safety profile for D.rubrovalvata mycelium. This makes it a promising candidate for further field trials.
