基于"成方-靶点-通路"的通络煎对脑梗死(气虚血瘀型)的作用机制

目的 探讨基于"成方-靶点-通路"的通络煎对脑梗死(气虚血瘀型)的作用机制。方法 从TCMSP及HERB数据库获得通络煎的成分和靶点,与GeneCards数据库中的脑梗死相关靶基因取交集,确定治疗靶点。利用STRING数据库在线构建治疗靶点蛋白互作网络。利用DAVID 6。8在线工具对治疗靶点进行GO富集分析和KEGG信号通路富集分析。结果 通络煎有492种有效成分、181个药物靶点,脑梗死有疾病靶点280个,药物疾病交集靶点29个,其中IL6、INS、PPARG、HIF1A、NOS3、VEGFA可能为核心治疗靶点。GO富集分析及KEGG信号通路富集分析显示,29个治疗靶点在60个生物学过程、23个细胞组分、13个分子功能、21条KEGG信号通路显著富集(P<0。01)。结论 通络煎通过多靶点、多通路防治脑梗死,具有改善脑循环、抑制缺血-再灌注损伤、改善神经功能缺损、抗动脉粥样硬化的作用。
Mechanism and clinical study of Tongluo decoction based on"formula target pathway"on cerebral infarction(Qi deficiency and blood stasis type)
Objective To investigate the mechanism of action of Tongluo decoction on cerebral infarction(Qi deficiency and blood stasis type)based on"formulation target pathway".Methods The components and targets of Tongluo decoction were obtained from TCMSP and HERB databases,and intersected with the target genes related to cerebral infarction in GeneCards database to determine the therapeutic targets.Constructing a therapeutic target protein interaction network online using the STRING database.Perform GO enrichment analysis and KEGG signaling pathway enrichment analysis on therapeutic targets using the DAVID 6.8 online tool.Results Tongluo decoction contains 492 active ingredients and 181 drug targets,with 280 disease targets for cerebral infarction and 29 drug disease intersection targets.Among them,IL6,INS,PPARG,HIF1A,NOS3,and VEGFA may be the core therapeutic targets.GO enrichment analysis and KEGG signaling pathway enrichment analysis showed that 29 therapeutic targets were significantly enriched in 60 biological processes,23 cellular components,13 molecular functions,and 21 KEGG signaling pathways(P<0.01).Conclusion Tongluojian can prevent and treat cerebral infarction through multiple targets and pathways,and has the effects of improving cerebral circulation,inhibiting ischemia-reperfusion injury,improving neurological deficit,and anti atherosclerosis.
