The effect of cell suspension concentration on the results of conventional culture micronucleus detection
Objective To explore a faster,unified,and effective method to improve the detection rate and film reading efficiency of micronuclei,and to provide experimental basis for the health examination of occupational radiation health personnel and other occupational exposed populations.Methods Human peripheral blood lymphocyte micronucleus test culture method:four different concentrations of cell suspensions(cell and supernatant volume ratios were 1∶1,1∶2,1∶3,1∶4,respectively)were set at the time of dropping,and micronucleus samples were prepared.The staining effect and morphological distribution after preparation were compared and analyzed under the microscope,and the film reading time was recorded to calculate the lymphocyte micronucleus rate and micronucleus cell rate.Results The results of microscopic examination showed that the staining effect and morphological distribution of micronucleus slides at concentrations of 1∶2 and 1∶3 were relatively ideal.There was a significant difference in the reading time under different concentrations(F=576.12,P<0.05),and the average reading time was ranked as 1∶4 concentration>1∶3 concentration>1∶2 concentration>1∶1 concentration.There was no significant difference in the number of micronucleus cells under different concentrations.The detection rate of micronucleus cells in micronucleus slides with a 1∶1 concentration was the lowest.Conclusion Producing high-quality micronucleus specimens can improve the detection rate and film reading efficiency of micronuclei.Through comparative analysis,it can be concluded that the production effect of cell suspension concentration of 1∶(2-3)is relatively ideal,which can improve the efficiency of film reading and the detection rate of micronuclei.
