Optimization extraction process and evaluation of antioxidant activity of polysaccharides from Trollius chinensis Bunge
Objective To optimize the extraction method of polysaccharides from Trollius chinensis Bunge and evaluate the antioxidant activity of them.Methods The polysaccharides from Trollius chinensis Bunge were extracted by hot water re-flux extraction method,and the effects of solid-liquid ratio,extraction temperature and extraction time on polysaccharides were investigated by a single factor test.On this basis,the orthogonal experimental design method was used to optimize the extraction conditions.The antioxidant activity of polysaccharides from Trollius chinensis Bunge were evaluated by measuring DPPH free radical,ABTS+free radical,hydroxyl free radical,superoxide anion free radical scavenging ability and total re-ducing power.Results The results showed that the optimal extraction process conditions of polysaccharides from Trollius chinensis Bunge were as follow:solid-liquid ratio of 1:25,extraction temperature of 80 ℃,and extraction time of 2 h.Under these conditions,the actual polysaccharides from Trollius chinensis Bunge was 1.350%±0.125%.The IC50 values of the scav-enging rates of DPPH free radical,ABTS+free radical,hydroxyl free radical and superoxide anion free radical were 0.161,0.079,0.006 and 4.216 mg·mL-1,respectively.While,when the concentration of polysaccharides was 4.0 mg·mL-1,the total reducing power was 2.78.Conclusion The optimized polysaccharides had an excellent antioxidant activity,which provided a theoretical basis for further exploitation and utilization of polysaccharides from Trollius chinensis Bunge.
