The influence of rapamycin on gap junction function and connexin expression
Objective To investigate the effect of rapamycin on gap junction(GJ)function and connexin(Cx)expres-sion in HUVEC,BRL-3A and AML12 cells.Methods CCK-8 assay was used to detect the cell viability of HUVEC,BRL-3A and AML12 cells treated with different concentrations of rapamycin.Parachute dye-coupling assay and Western blotting were used to detect GJ function and Cx expression level.Results With rapamycin at 5,10,25,50 nmol·L-1 concentrations for 2 h,the cell growth of HUVEC,BRL-3A and AML12 were not affected,and the dye coupling were reduced comparing with control(P<0.05),and the expression level of Cx37 and Cx32 were significantly decreased at concentrations of 25 and 50 nmol·L-1(P<0.05).Conclusion Rapamycin could inhibit the GJ function and downregulate the expression of Cx37 and Cx32 in HUVEC,BRL-3A and AML12 cells.
