Analysis on the expression and correlation of Cullin7 in gastric cancer
Objective To investigate the expression of Cullin7 in gastric cancer and its role in the genesis and devel-opment of gastric cancer.Methods Download the mRNA level data of gastric cancer patients from The Cancer Genome At-las(TCGA)database,and analyze the survival status;The expression level of Cullin 7 in gastric cancer and adjacent tissues of 60 patients with gastric cancer undergoing surgical treatment was analyzed by immunohistochemistry,and the correlation between the expression level of Cullin 7 and the clinicopathological characteristics of gastric cancer patients was evaluated.At the same time,through overexpression or knockout of Cullin7 gene in human gastric cancer HGC-27 cell line,the effects of Cullin7 on the proliferation,migration and invasion of gastric cancer cells were studied by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT),colony formation,wound healing,Transwell migration and Matrigel invasion experiments.Results Cullin7 is highly expressed in gastric cancer,and patients with high expression of Cullin7 have shorter overall survival,and shorter first progression and post progression survival times.At the same time,the expression of Cullin7 is significantly related to the pathologic differentiation,nerve invasion,vascular invasion,depth of invasion,lymph node metastasis and clinical stage of gastric cancer patients.The ectopic expression of Cullin7 in gastric cancer cells signifi-cantly promotes cell proliferation,migration and invasion,while knocking down Cullin7 with short hairpin RNA(shRNA)can inhibit the proliferation,migration and invasion of gastric cancer cells.Conclusion Upregulation of Cullin7 may be an important factor promoting the occurrence and development of gastric cancer,and can be used as a new biomarker for gastric cancer diagnosis and prognosis as well as a new therapeutic target.
