Extraction,purification and structural analysis of polysaccharides from Fuscoporia punctata(Fr.)Cunn
Objective To analyze and characterize the structure of BHP90-3L,a homogeneous acidic polysaccharide isolated and purified from Fuscoporia punctata(Fr.)Cunn.Methods Using DEAE Sepharose,Sephadex G-75,and other column chromatography techniques,the polysaccharides of F.punctata(Fr.)Cunn were isolated and purified.The absolute molecular weight(Mw),monosaccharide composition,type of glycosidic linkage,and conformation of the isolated acidic polysaccharide BHP90-3L from F.punctata(Fr.)Cunn were investigated to determine its structure through monosaccharide composition analysis,infrared analysis,methylation analysis,and NMR spectroscopy data analysis.Results BHP90-3L is an acidic heteropolysaccharide with a molecular weight of 9.489×103 Da,mainly containing mannose,glucuronic acid,glucose,and galactose,with a molar ratio of 2.78∶1.72∶7.17∶1.00 for the monosaccharide composition.From the GC-MS and NMR results,it is hypothesized that the main chain of BHP90-3L is composed of→6)-α-D-Manp-(1→,→3)-β-D-Glcp-(1→,→3)-β-D-Glcp A-(1→,→3,4)-α-D-Manp-(1→,→6)-α-D-Manp-(1→.Among them,→3)-β-D-Glcp-(1→ and →3)-β-D-Glcp A-(1→)both have repeating units,and the side chain has only β-D-Galp-(1→.The Congo red experiment indicates that BHP90-3L does not have a three-stranded helical structure.Conclusion BHP90-3L,a new homogeneous acidic polysaccharide,was isolated from F.punctata(Fr.)Cunn.The study of its structure provides a reference for the further development and utilization of F.punctata(Fr.)Cunn polysaccharides.
