Effects of medicated serum of Isodon ternifolius(D.Don)Kudo on TLR4/NF-κB/NLRP3 signaling pathway in hepatocytes
Objective To investigate the effect of medicated serum of Isodon ternifolius on TLR4/NF-κB/NLRP3 in primary rat hepatocyte model.Methods Hepatocytes induced by LPS were divided into 7 groups from A to G.The cell proliferation was detected by MTT assay;The biochemical methods were employed to determine the levels of ALT and AST in hepatocytes;ELISA was used to measure IL-1β,IL-18 and caspase-1;RT-PCR,WB and immunofluorescence analysis were used to detect the expression of TLR4,p-IκB-α,NF-κB,IκB-α,NLRP3,GSDMD and ASC expression.Results The results indicate that the proliferation of hepatocytes was impeded,resulting in a suppression of the secretion of ALT,AST,caspase-1,IL-1β and IL-18.Furthermore,the expression of TLR4,NF-κB p65,NLRP3,ASC,and GSDMD was notably diminished,while the expression of IκB-α was significantly upregulated in the medicated serum derived from Isodon ternifolius.Conclusion The medicated serum of Isodon ternifolius can prevent hepatocyte activation by inhibiting TLR4/NF-κB/NLRP3 signaling,down-regulate the release of hepatocyte inflammatory factors,and reduce hepatocyte inflammatory damage.
