Effects of different strains and phenylalanine supplementation on accumulation of flavonoids in Spirodela polyrhiza(L.)Schleid.
Objective To explore the main flavonoid constituents across various strains of Spirodela polyrhiza(L.)Schleid.,and to assess the impact of phenylalanine supplementation on the growth and total flavonoid yield of these strains,the following investigation was conducted.Methods Morphological and DNA barcoding identification were performed on duckweed samples collected from Tianjin,Wuhan,and Guiyang.Luteolin,orientin and vitexin levels in six strains were quan-tified using UPLC.Different S.polyrrhiza strains were cultivated with 0,0.005,0.05,0.5 and 1 mmol·L-1 phenylalanine for 10 days,followed by measurement of fresh weight,dry weight and total flavonoid contents.Results The collected duckweed samples were identified as Spirodela.Strain p143 exhibited the highest total flavonoids and three main flavonoids contents a-mong the six strains.Phenylalanine supplementation at 0.005 mmol·L-1 significantly enhanced fresh weight accumulation in strains WHD and 5543.Treatment with 0.05 mmol·L-1 phenylalanine increased the total flavonoid content by 16.5%,33.1%and 42.6%in strains 5543,W0716 and 9509,respectively.Conclusion Strain p143 exhibited the highest flavonoid content among the six strains.Low-dose phenylalanine supplementation can promote the growth and total flavonoid contents accumu-lation of some strains of S.polyrrhiza.
