Molecular Marker-Assisted Selection of Germplasm Resources Resistant to Potato Virus Y from Potato Wild Relatives
To broaden the genetic bases of tetraploid potato(2n=4x=48)with the resistance to potato virus Y(PVY),56 accessions of potato wild relatives were screened by anti-PVY molecular markers(RY-SC3,YES3-3A and Ry364).The PCR results showed that thirteen accessions carried the marker RYSC3 link-ing to Ryad gresistant gene(originally found in Solanum tuberosum ssp.andigena),which included 3 dihaploid(2n=2x=24)accessions,6 tetraploid accessions and 4 hexaploid(2n=6x=72)accessions.Four accessions carried the marker YES3-3A linking to Rysto resistant gene(originally found in S.stoloniferum Schlechtdal),which included 1 dihaploid accession,2 tetraploid accessions and 1 ploidy-undetermined accession.Twenty three accessions carried the marker Ry364 linking to Rychc resistant gene(originally found in S.chacoense Bit-ter),which included 6 dihaploid accessions,4 tetraploid accessions and 13 hexaploid accessions.Seven ac-cessions carried 2 different molecular markers among RYSC3,YES3-3A and Ry364,while only 1 accession,OCH 14180a(S.stoloniferum Schlechtdal)carried all the 3 markers.These selected accessions could be used for the anti-virus breeding,germplasm innovation and gene mining.
