Differential Analysis of Volatile Metabolites in Flue-Cured Tobacco Leaves Under Different Spraying Methods of Exogenous Enzyme at Initial Curing Stage
In order to clarify the variation rules of differential volatile metabolites in flue-cured tobacco leaves under different spraying methods of exogenous enzyme preparations at the initial curing stage(yelting stage),Yunyan 99(B2F)from Luonan County,Shangluo City,Shaanxi Province was used as test material in this experiment,non-targeted metabolomics analysis was performed by liquid chromatography-mass spectrome-try(LC-MS),and the differences in volatile metabolites of flue-cured tobacco leaves under different enzyme spraying methods(LN and LN-A)and the control(LN-CK)were studied.The results showed that the applica-tion methods of exogenous enzyme had significant influences on the relative content of volatile metabolites in tobacco leaves.A total of 578 volatile metabolites were detected from tobacco leaves,including terpenoids(99 kinds),esters(91 kinds),heterocyclic compounds(88 kinds),hydrocarbons(63 kinds),ketones(52 kinds),aldehydes(42 kinds),organic acids(21 kinds)and so on.The results of PCA and OPLS-DA analy-sis showed that the metabolites between LN-CK and LN,LN-CK and LN-A,and LN and LN-A separated well,and 177,121 and 175 kinds significantly different metabolites were observed,respectively.Among which,142 kinds showed increase in relative content and 35 kinds showed decrease in LN treatment compared with LN-CK;71 kinds showed increase and 50 kinds showed decrease in LN-A treatment compared with LN-CK;56 kinds showed increase and 119 kinds showed decrease in LN-A treatment compared with LN treatment.Based on the S-plot and volcano plot between samples,20 volatile components with significant differences in relative content were screened out from each group of LN-CK and LN,LN-CK and LN-A,and LN and LN-A,so that the different effects of the two enzyme spraying treatments could be compared with them.It could pro-vide technical support for the selection of exogenous enzyme spraying methods.All in all,the spraying method of exogenous enzyme preparations in LN treatment could better positively affect the types of volatile metabolites and improve the aroma quality of flue-cured tobacco leaves.
