小菜蛾幼虫生防菌株毒力测定及产毒条件优化

为丰富小菜蛾(Plutella xylostella)生物防治的微生物资源,本研究选用粉红粘帚霉(Clonostachys rosea)、淡紫拟青霉(Purpureocillium lilacinum)和 02 号生赤壳属(Bionectria sp。)三种具有较好生防效果的菌株开展其对小菜蛾幼虫毒力的菌株筛选试验,同时对选出的高毒力菌株进行毒力测定和产毒条件优化。结果表明,三种菌株对小菜蛾幼虫都具有防治效果,其中以粉红粘帚霉的生防毒力最强。毒力测定显示,粉红粘帚霉的半致死时间(LT50)为30。88 h,半致死浓度(LC50)为4。365×104个孢子/mL。从产毒条件优化结果看出,粉红粘帚霉产毒的最适培养基为萨氏葡萄糖琼脂酵母膏(SDAY),最适pH值为 6,最适培养时间为 7d。综上表明,粉红粘帚霉对小菜蛾幼虫具有毒力高、致病速度快等特点,具有较大的生防潜力,可作为防治小菜蛾的优良生防真菌。
Virulence Determination and Optimization of Toxin Production Conditions of Biocontrol Strains of Plutella xylostella Larvae
To enrich microbial resources for biological control of Plutella xylostella,this study was con-ducted using three fungi strains as Clonostachys rosea,Purpureocillium lilacinum and Bionectria sp.02 with good biocontrol effects.Their virulence to the larvae of P.xylostella was tested,and the strain with high viru-lence was selected,then its virulence was determined and the toxin production conditions were optimized.The results showed that the three strains were all effective in controlling the larvae of P.xylostella,and the most ef-fective one was C.srosea.The virulence test showed that the half lethal time(LT50)and half lethal concentra-tion(LC50)of C.rosea were 30.88 h and 4.365×104 spores per milliliter.According to the optimization results of toxin production conditions of C.rosea,the optimal medium was SDAY,the optimal pH value was 6,and the optimal culture time was 7 days.In summary,C.rosea had the characteristics of high virulence and rapid pathogenicity to the larvae of P.xylostella,so it had great biocontrol potential and could be used as an excel-lent biocontrol fungus against P.xylostella.
