Identification and Screening of Salt Tolerant Germplasm Resources of Sorghum at Germination Stage
Germination stage is the most sensitive period to saline stress in sorghum growth.Identifying and screening salt tolerant sorghum germplasms at germination stage is of great significance for breeding salt tolerant sorghum varieties.The current study used 150 mmol/L NaCl to simulate salt stress environment,and the salt tolerance of 172 sorghum germplasms during germination were evaluated.The relative values of nine indicator as germination potential,germination rate,germination index,bud length,root length,bud fresh weight,bud dry weight,root fresh weight and root dry weight were calculated for correlation analysis,princi-pal component analysis and cluster analysis.The results demonstrated that the 172 sorghum germplasms had a-bundant genetic variability in these indicators,and significantly or very significantly positive correlations were found among most of these indicators.Through principal component analysis,the original nine indicators were summarized into three principal components representing germination status and bud and root growth status,and among which,germination index,dry weight of buds and root length could be used as main indicators for identifying salt tolerance of sorghum at germination stage.The results of comprehensive evaluation by member-ship function method and cluster analysis showed that the 172 sorghum germplasms were divided into five groups,which included 34 highly salt-tolerant germplasms,35 salt tolerant germplasms,35 medium salt-toler-ant germplasms,35 salt-sensitive germplasms,and 31 highly salt-sensitive germplasms,respectively.Among them,R4545,TX622B and R366 had the strongest salt tolerance at germination stage,which could provide the materials for salt tolerant variety breeding,genetic improvement and molecular mechanisms studies of salt tolerant sorghum in the next step.
