6株核桃腐烂病菌的分子鉴定及生物学特性比较

为探究核桃腐烂病菌的分类地位及生物学特性,本研究基于分子生物学方法对 6 株菌落形态有差异的核桃腐烂病菌进行多基因联合鉴定,并进一步探究其致病性及生物学特性的差异。系统进化树显示,44-9-2B与Cytospora melnikii聚为一支,且自举支持率为 100%;菌株 44-1-2C、51-1-3C与C。chrsosperma聚为一支;菌株 49-2-1A、49-9-4D、53-1-3F与Valsa sordida聚为一支。致病性检测表明,44-9-2B致病性最强,也最先产生分生孢子角,其次为菌株 44-1-2C和 51-1-3C,最后是菌株 53-1-3F。生物学特性研究表明,6 株致病菌最适生长温度均为 28℃,最适pH值为 6,光照对 6 株致病菌的生长有促进作用,各致病菌的最适氮、碳源不同。该研究结果可为核桃树腐烂病的防治提供理论依据。
Molecular Identification and Comparison on Biological Characteristics of Six Walnut Canker Pathogens
In order to investigate the specific types and biological characteristics of walnut canker patho-gens,six isolates of walnut canker pathogen with discrepant colony morphologies were jointly identified with multiple genes based on molecular biological methods,and the variations in their pathogenicity and biological characteristics were further explored.The results indicated that based on the phylogenetic tree,the strain 44-9-2B was clustered with Cytospora melnikii with the bootstrap support rate of 100%;the strains 44-1-2C and 51-1-3C were clustered with C.chrsosperma;the strains 49-2-1A,49-9-4D and 53-1-3F were clustered with Valsa sordida.The strain 44-9-2B had the strongest pathogenicity and produced conidia first,followed by strains 44-1-2C and 51-1-3C,while strains 49-2-1A,49-9-4D and 53-1-3F were the last.The results of biological char-acteristics showed that the optimal growth temperature for the six pathogenic strains was 28℃,and the optimal pH value was 6;light had promoting effects on the growth of the six pathogenic strains;but the optimal nitrogen and carbon sources were different for different pathogenic strains.The findings of this study could provide a theoretical basis for prevention and control of the walnut canker pathogens.
