Expression changes of miR-331-3p and MLLT10 mRNA in colorectal cancer and their clinical significance
Objective To observe expression changes of microRNA-331-3P(miR-331-3p)and mixed-lineage leuke-mia transmutation cofactor 10(MLLT10)mRNA in the colorectal cancer(CRC)tissues,and to explore the relationships between the expression and clinicopathological features and epithelial-mesenchymal transition(EMT).Methods Totally 156 patients with CRC were selected,and CRC tissues and their matched paracancer normal tissues were obtained during operation.Real-time fluorescence quantitative PCR was used to detect the mRNA expression levels of miR-331-3p,MLLT10 and EMT-related genes E-cadherin,N-cadherin and Snail.The mRNA expression levels of miR-331-3p,MLLT10,E-cadherin,N-cadherin and Snail were compared between the CRC and paracancer normal tissues.The relation-ships between the mRNA expression of miR-331-3p and MLLT10 and clinicopathological features,and the relationships between the expression of miR-331-3P and MLLT10 and the mRNA expression of E-cadherin,N-cadherin and Snail in CRC were analyzed.Results The relative expression levels of MLLT10,N-cadherin and Snail mRNA in the CRC were higher than those in the paracancer normal tissues,and the relative expression levels of miR-331-3p and E-cadherin mRNA were lower than those in the paracancer normal tissues(all P<0.01).The mRNA expression levels of miR-331-3p and MLLT10 in CRC tissues were related to TNM stage and lymph node metastasis(all P<0.05),but not to gender,age,maximum tumor diameter,tumor location,or tissue differentiation degree(all P>0.05).Pearson correlation analysis showed that the expression of miR-331-3p in CRC tissues was negatively correlated with MLLT10 mRNA expression(r=-0.678,P<0.05).The expression of miR-331-3p in CRC tissues was positively correlated with the mRNA expression of E-cadherin(r=0.589,P<0.05),and was negatively correlated with the mRNA expression levels of N-cadherin and Snail(r=-0.712 and-0.654,respectively;all P<0.05).The mRNA expression of MLLT10 in CRC was negatively correlated with the mRNA expression of E-cadherin(r=-0.549,P<0.05),and was positively correlated with the mRNA expression levels of N-cadherin and Snail(r=0.668 and 0.714,respectively;all P<0.05).Conclusion Low expression of miR-331-3p and high expression of MLLT10 mRNA in CRC are closely related to TNM staging,lymph node metastasis,and EMT.
