目的 观察结直肠癌(CRC)组织微小RNA-331-3p(miR-331-3p)、混合谱系白血病转位辅因子 10(MLLT10)mRNA表达变化,并探讨其表达与临床病理特征和上皮间质转化的关系。方法 选择CRC患者156例,取术中获取的CRC组织及其配对的癌旁正常组织,采用实时荧光定量聚合酶链反应检测miR-331-3p、MLLT10 mRNA表达以及上皮间质转化相关基因E-钙黏蛋白(E-cadherin)、N-钙黏蛋白(N-cadherin)、锌指转录因子Snail mRNA表达。比较CRC组织与癌旁正常组织miR-331-3p及MLLT10、E-cadherin、N-cadherin、Snail mRNA表达;分析CRC组织miR-331-3p、MLLT10 mRNA表达与临床病理特征的关系,二者表达的关系及其与E-cadherin、N-cadherin、Snail mRNA表达的关系。结果 CRC组织MLLT10、N-cadherin、Snail mRNA相对表达量均高于癌旁正常组织,miR-331-3p、E-cadherin mRNA相对表达量均低于癌旁正常组织(P均<0。01)。CRC组织miR-331-3p、MLLT10 mRNA表达与TNM分期、淋巴结转移有关(P均<0。05),而与性别、年龄、肿瘤最大径、肿瘤位置、组织分化程度无关(P均>0。05)。Pearson相关分析显示,CRC组织miR-331-3p表达与MLLT10 mRNA表达呈负相关(r=-0。678,P<0。05);CRC组织miR-331-3p表达与E-cadherin mRNA表达呈正相关(r=0。589,P<0。05),与N-cadherin、Snail mRNA表达呈负相关(r分别为-0。712、-0。654,P均<0。05);CRC组织MLLT10 mRNA表达与E-cadherin mRNA表达呈负相关(r=-0。549,P<0。05),与N-cadherin、Snail mRNA表达呈正相关(r分别为0。668、0。714,P均<0。05)。结论 CRC组织miR-331-3p低表达、MLLT10 mRNA高表达,二者表达与TNM分期、淋巴结转移以及上皮间质转化密切相关。
Expression changes of miR-331-3p and MLLT10 mRNA in colorectal cancer and their clinical significance
Objective To observe expression changes of microRNA-331-3P(miR-331-3p)and mixed-lineage leuke-mia transmutation cofactor 10(MLLT10)mRNA in the colorectal cancer(CRC)tissues,and to explore the relationships between the expression and clinicopathological features and epithelial-mesenchymal transition(EMT).Methods Totally 156 patients with CRC were selected,and CRC tissues and their matched paracancer normal tissues were obtained during operation.Real-time fluorescence quantitative PCR was used to detect the mRNA expression levels of miR-331-3p,MLLT10 and EMT-related genes E-cadherin,N-cadherin and Snail.The mRNA expression levels of miR-331-3p,MLLT10,E-cadherin,N-cadherin and Snail were compared between the CRC and paracancer normal tissues.The relation-ships between the mRNA expression of miR-331-3p and MLLT10 and clinicopathological features,and the relationships between the expression of miR-331-3P and MLLT10 and the mRNA expression of E-cadherin,N-cadherin and Snail in CRC were analyzed.Results The relative expression levels of MLLT10,N-cadherin and Snail mRNA in the CRC were higher than those in the paracancer normal tissues,and the relative expression levels of miR-331-3p and E-cadherin mRNA were lower than those in the paracancer normal tissues(all P<0.01).The mRNA expression levels of miR-331-3p and MLLT10 in CRC tissues were related to TNM stage and lymph node metastasis(all P<0.05),but not to gender,age,maximum tumor diameter,tumor location,or tissue differentiation degree(all P>0.05).Pearson correlation analysis showed that the expression of miR-331-3p in CRC tissues was negatively correlated with MLLT10 mRNA expression(r=-0.678,P<0.05).The expression of miR-331-3p in CRC tissues was positively correlated with the mRNA expression of E-cadherin(r=0.589,P<0.05),and was negatively correlated with the mRNA expression levels of N-cadherin and Snail(r=-0.712 and-0.654,respectively;all P<0.05).The mRNA expression of MLLT10 in CRC was negatively correlated with the mRNA expression of E-cadherin(r=-0.549,P<0.05),and was positively correlated with the mRNA expression levels of N-cadherin and Snail(r=0.668 and 0.714,respectively;all P<0.05).Conclusion Low expression of miR-331-3p and high expression of MLLT10 mRNA in CRC are closely related to TNM staging,lymph node metastasis,and EMT.
