miR-498过表达对鼻咽癌细胞增殖、侵袭、迁移和上皮间质转化的影响及其机制

目的 探讨微小RNA-498(miR-498)过表达对鼻咽癌细胞增殖、侵袭、迁移和上皮间质转化(EMT)的影响及其机制。方法 体外传代培养人永生化鼻咽上皮细胞系NP69及人鼻咽癌细胞系CNE-2Z、CNE-1、HNE-1、SUNE-1,采用RT-qPCR法检测各细胞miR-498表达,选择miR-498表达最低的鼻咽癌细胞进行后续实验。取传3代、对数生长期、生长状态良好的鼻咽癌细胞,随机分为miR-498 mimics组和NC mimics组,分别转染miR-498 mimics、NC mimics。转染6 h更换新鲜培养基继续培养24 h,收集细胞。采用CCK-8法检测细胞增殖活性,采用平板克隆形成实验检测集落形成能力,采用Transwell小室法检测细胞侵袭和迁移能力,采用Western blotting法检测EMT相关上皮型钙黏素(E-cadherin)、波形蛋白(Vimentin)、纤连蛋白(Fibronectin)以及高迁移率族蛋白A2(HMGA2)表达。结果 CNE-1、HNE-1、CNE-2Z、SUNE-1细胞miR-498相对表达量均低于NP69细胞(P均<0。05)。以CNE-2Z细胞miR-498相对表达量最低,故选择该细胞系进行后续实验。经验证,miR-498 mimics组miR-498相对表达量高于NC mimics组(P<0。05)。miR-498 mimics组培养0、24、48、72 h细胞增殖活性均低于NC mimics组(P均<0。05);miR-498 mimics组集落形成能力以及细胞侵袭和迁移能力均低于NC mimics组(P均<0。05);miR-498 mimics组Vimentin、Fibronectin、HMGA2蛋白相对表达量均低于NC mimics组,E-cadherin蛋白相对表达量高于NC mimics组(P均<0。05)。结论 鼻咽癌细胞miR-498低表达;miR-498过表达则能抑制鼻咽癌细胞的增殖、侵袭、迁移以及EMT,其机制可能与靶向调控HMGA2表达有关。
Effects of overexpression of miR-498 on proliferation,invasion,migration,and EMT of nasopharyngeal carcinoma cells
Objective To investigate the effects of overexpression of microRNA-498(miR-498)on proliferation,invasion,migration,and epithelial-mesenchymal transition(EMT)of nasopharyngeal carcinoma cells and their mecha-nisms.Methods Human immortalized nasopharyngeal epithelial cells NP69 and human nasopharyngeal carcinoma cell lines CNE-2Z,CNE-1,HNE-1,SUNE-1 were cultured in vitro.RT-qPCR was used to detect the relative expression of miR-498 in each cell line,and nasopharyngeal carcinoma cells with relatively low miR-498 expression were selected for subsequent experiments.Nasopharyngeal cancer cells in the 3rd generation,which were in logarithmic growth phase and had good growth status,were randomly divided into the miR-498 mimics group and NC mimics group,and were transfected with miR-498 mimics and NC mimics,respectively.After 6 hours of transfection,the medium was replaced with fresh cul-ture medium and the culture continued for 24 h.Cell activity was detected using the CCK-8,colony formation ability was detected using plate cloning assay,cell invasion and migration abilities were detected using Transwell cell assay,and the expression of EMT-related epithelial cadherin(E-cadherin),Vimentin,Fibronectin,and high-mobility group protein A2(HGMA2)was detected by Western blotting.Results The relative expression levels of miR-498 in the human nasopha-ryngeal carcinoma cells CNE-1,HNE-1,CNE-2Z,and SUNE-1 were lower than that in NP69 cells(all P<0.05).In the nasopharyngeal carcinoma cells,the relative expression level of miR-498 in CNE-2Z cells was relatively low,so CNE-2Z cells were selected for subsequent experiments.After verification,the relative expression level of miR-498 in the miR-498 mimics group was higher than that in the NC mimics group(P<0.05).The proliferation activity of cells in the miR-498 mimics group at 0,24,48,and 72 h of culture was lower than that in the NC mimics group(P<0.05).The colony forma-tion,invasion and migration abilities of miR-498 mimics group were lower than those of the NC mimics group(all P<0.05).The relative expression levels of Vimentin,Fibronectin,and HMGA2 proteins in the miR-498 mimics group were lower than those in the NC mimics group,while the relative expression level of E-Cadherin protein was higher than that in the NC mimics group(all P<0.05).Conclusion The expression of miR-498 in nasopharyngeal carcinoma cells is low;overexpression of miR-498 can inhibit the proliferation,invasion,migration,and EMT of nasopharyngeal carcinoma cells,and its mechanism may be related to targeted regulation of HMGA2.
