Effects of herbal-cake-separated moxibustion on co-stimulatory molecules CTLA-4,4-1BB and CD28 in immunosuppression rabbits induced by cyclophosphamide
Objective To investigate the effects of herbal-cake-separated moxibustion on co-stimulatory molecules cytotoxic T-lymphocyte-associated antigen 4(CTLA-4),4-1BB and CD28 in immunosuppressive rabbits induced by cyclo-phosphamide.Methods Thirty-two big-eared white rabbits were randomly divided into the blank group,model group,moxibustion group and herbal-cake-separated moxibustion group,with 8 rabbits in each group.Rabbits in the model group,moxibustion group and herbal cake-separated moxibustion group were given cyclophosphamide to induce the immu-nosuppression models.The intervention of moxibustion and herbal-cake-separated moxibustion started on the 2nd day after successful modeling in the moxibustion group and herbal-cake-separated moxibustion group,once every other day,a total of 10 times.No moxibustion was given in the blank group and model group.On the day after the last intervention,the abdominal venous blood was collected in the moxibustion group and moxibustion group as well as in the blank group and model group at the same time.The serum was retained by centrifugation,and serum CTLA4,4-1BB and CD28 were detected by ELISA.After abdominal vein blood collection,spleen and liver were extracted,and the expression levels of CTLA-4 and 4-1BB in spleen and liver tissues were detected by immunohistochemistry.Results The serum levels of CTLA-4 and CD28 in the model group were higher than those in the blank group,and the level of 4-1BB was lower than that in the blank group(all P<0.05).Compared with the model group,the serum levels of CTLA-4 and CD28 decreased and the serum level of 4-1BB increased in the moxibustion group and the herbal-cake-separated moxibustion group(all P<0.05).Compared with the moxibustion group,the serum CTLA-4 and CD28 levels decreased and the serum 4-1BB level increased in the herbal-cake-separated moxibustion group(all P<0.05).The positive expression of CTLA-4 in spleen and liver tissues in the model group was higher than that in the blank group,and the positive expression of 4-1BB in spleen and liver tissues was lower than that in the blank group(all P<0.05).Compared with the model group,the positive expression of CTLA-4 in spleen and liver tissues decreased in the moxibustion group(P<0.05),the positive expression of 4-1BB in liver tissue was elevated(P<0.05),and the increase of the positive expression of 4-1BB in spleen tissue was not significant(P>0.05).The positive expression of CTLA-4 in spleen and liver tissues decreased(P<0.05),and the positive expression of 4-1BB in spleen and liver tissues increased in the herbal-cake-separated moxibustion group(P<0.05).There was no sig-nificant difference in the positive expression levels of CTLA-4 or 4-1BB in spleen and liver tissues between the moxibustion group and the herbal-cake-separated moxibustion group(both P>0.05).Conclusion Herbal-cake-separated moxibus-tion can improve immune function by regulating co-stimulatory molecules CTLA-4,4-1BB and CD28 of immunosuppres-sive rabbit,and its effect is better than that of moxibustion.
