Effect of Aidi injection on radiosensitivity of cervical cancer cells
Objective To study the effect and mechanism of Aidi injection on radiosensitivity of cervical cancer cells.Methods SiHa cells from cervical cancer were cultured and randomly divided into the blank control group and Aidi injection group.Cells in the blank control group were added with 100 μL of cell culture media,and cells in the Aidi injection group were added with 100 μL of 10 mg/mL Aidi injection.After 24 h of culture,the cells in the two groups were subjected to radiation therapy using the medical linear accelerator at doses of 0,2,4,6,and 8 Gy,respectively.We ob-served the plating efficiency(PE)of two groups at 0 Gy,and calculated the survival fraction(SF)of the two groups at 2,4,6,and 8 Gy based on PE.A decrease in SF indicated an increase in radiation sensitivity.SiHa cells were divided into the blank control group,radiotherapy group,and Aidi+radiotherapy group.Cells in the blank control group were added with 100 μL of cell culture medium.After adding 100 μL of cell culture media,the radiotherapy group received a radia-tion dose of 10 Gy.After adding 100 μL of 10 mg/mL Aidi injection,the Aidi+radiotherapy group received a radiation dose of 10 Gy.Flow cytometry was used to observe the apoptosis of three groups of cells,and real-time quantitative PCR was applied to detect the expression of B-cell lymphoma-2(Bcl-2)mRNA in cells.Results The SF of the Aidi injection group at irradiation doses of 2,4,6,and 8 Gy was significantly lower than that of the blank control group(P=0.002).The apoptosis rate of the radiotherapy group was significantly higher than that of the blank control group,and the apoptosis rate of the Aidi+radiotherapy group was significantly higher than that of the radiotherapy group(all P<0.01).The expres-sion of Bcl-2 mRNA in the radiotherapy group was significantly lower than that in the blank control group,while the expres-sion of Bcl-2 mRNA in the Aidi+radiotherapy group was significantly lower than that in the radiotherapy group(both P<0.01).Conclusion Aidi injection can significantly increase the radiosensitivity of cervical cancer SiHa cells,and its mechanism may be to promoting apoptosis by inhibiting the expression of Bcl-2.
