Expression of GPT2 in ESCC tissues and its effects on cell proliferation,migration and invasion
Objective To observe the expression of glutamic pyruvate transaminase 2(GPT2)in the esophageal squa-mous cell carcinoma(ESCC)tissues,and to analyze the relationships between GPT2 and the clinicopathological features and prognosis of patients,as well as its effects on the proliferation,migration and invasion of ESCC cells in order to explore its relationship with progression of ESCC.Methods Eighty cases of ESCC tumor tissues and adjacent normal tissues were selected,and 3 of them were screened for differentially expressed genes by transcriptome sequencing.The lowly expressed gene GPT2 was identified as the research object,and its expression in ESCC was verified in TCGA database.Western blot-ting and immunohistochemistry were used to detect the protein levels of GPT2 in ESCC tissues,respectively.Multivariate Cox survival risk analysis was used to determine the risk factors affecting the survival of ESCC patients,and the relation-ship between GPT2 expression and clinicopathological characteristics of patients was explored.Kaplan-Meier survival anal-ysis was used to analyze the survival of patients with high or low GPT2 expression.The pIRES2/GPT2(GPT2 overexpres-sion)plasmid was constructed,and then the empty plasmid(pIRES2)and pIRES2/GPT2 were transfected into human esophageal squamous cell lines KYSE150 and KYSE30(transfected cells were named pIRES2 and pIRES2/GPT2 groups,respectively).After 48 h of culture,the proliferation,migration and invasion abilities of ESCC cells were observed by CCK-8,and Transwell assay.Results Transcriptome sequencing results showed that the expression level of GPT2 de-creased in ESCC tissues in comparison with that of the adjacent normal tissues(P<0.05).Western blotting and immuno-histochemistry confirmed that GPT2 protein levels were markedly down-regulated in the ESCC tissues(both P<0.05).Multivariate Cox survival risk analysis showed that lymph node metastasis,pathological stage and low expression of GPT2 were the survival risk factors of ESCC patients(all P<0.05),and GPT2 expression was notably related to lymph node me-tastasis(P<0.05).Survival analysis showed that the survival rate of patients with low GPT2 expression was significantly lower than that of patients with high GPT2 expression(P<0.05).The GPT2 overexpression plasmid was successfully con-structed and verified.Overexpression of GPT2 markedly inhibited the proliferation,migration and invasion of ESCC cells(all P<0.05).Conclusions The expression of GPT2 was down-regulated in the ESCC tissues,and the survival progno-sis of patients with low expression of GPT2 is poor.Overexpression of GPT2 can inhibit the proliferation,migration and in-vasion of ESCC cells.
