鸦胆子油乳对肺癌A549细胞顺铂化疗的增敏作用及其机制

目的 观察鸦胆子油乳对肺癌A549细胞顺铂化疗的增敏作用并分析其机制。方法 体外培养人肺癌细胞A549,MTT法检测鸦胆子油乳、顺铂对细胞生长的抑制率,计算鸦胆子油乳、顺铂对细胞的半数抑制浓度(IC50),观察不同浓度(2。44、9。77、78。12 μg/mL)鸦胆子油乳对顺铂IC50的影响。将A549细胞分为对照组、鸦胆子油乳组、顺铂组及鸦胆子油乳+顺铂组,对照组不做处理正常培养细胞,鸦胆子油乳组、顺铂组及鸦胆子油乳+顺铂组分别给予鸦胆子油乳单药、顺铂单药、鸦胆子油乳及顺铂联合处理。采用荧光显微镜观察细胞形态及生长情况,流式细胞术检测细胞周期及细胞凋亡率,二氯荧光素二乙酸酯荧光探针染色检测细胞内ROS水平。结果 顺铂IC50随着鸦胆子油乳处理浓度的增高而下降(P均<0。05)。对照组细胞生长较好,细胞数量较多且形状为正常生长状态;顺铂组及鸦胆子油乳组细胞数量较对照组均减少,可见少量碎片;鸦胆子油乳+顺铂组细胞生长受抑制较明显,细胞数量最少,细胞形状不规则且细胞碎片增多。G0/G1期细胞比例鸦胆子油乳+顺铂组>顺铂组、鸦胆子油乳组>对照组,细胞凋亡率鸦胆子油乳+顺铂组>鸦胆子油乳组、顺铂组>对照组,细胞内ROS水平鸦胆子油乳+顺铂组>鸦胆子油乳组、顺铂组>对照组(P均<0。05)。结论 鸦胆子油乳能够增加肺癌A549细胞对顺铂化疗的敏感性,其机制可能与阻滞肿瘤细胞于G0/G1期、增加活性氧产生从而促进细胞凋亡有关
Sensitization effect and mechanism of Brucea Javanica oil emulsion on cisplatin chemotherapy in lung cancer A549 cells
Objective To observe the sensitization effect of Brucea Javanica oil emulsion on cisplatin chemotherapy in lung cancer A549 cells and to analyse its mechanism.Methods Human lung cancer cells A549 were cultured in vi-tro.MTT method was used to detect the inhibitory rates of Brucea Javanica oil emulsion,cisplatin,Brucea Javanica oil emulsion combined with cisplatin on cell growth.The half inhibitory concentration(IC50)of Brucea Javanica oil emulsion and cisplatin on cells was calculated,and the effects of different concentrations of Brucea Javanica oil emulsion(2.44,9.77,78.12 μg/mL)on cisplatin IC50 were observed.A549 cells were divided into four groups:the control group,Brucea Javanica oil emulsion group,cisplatin group,and Brucea Javanica oil emulsion + cisplatin group.Cells in the control group were routinely cultured,while cells in the Brucea Javanica oil emulsion group,cisplatin group,and Brucea Javanica oil emulsion+cisplatin group were treated with Javanica oil emulsion,cisplatin,Brucea Javanica oil emulsion and cisplat-in,respectively.Fluorescence microscopy was used to observe cell morphology and growth,flow cytometry was used to de-tect cell cycle and apoptosis rate,and dichlorofluorescein diacetate fluorescent probe staining was used to detect intracellu-lar reactive oxygen species(ROS)levels.Results The IC50 of cisplatin decreased with the increase of the concentrations of Brucea Javanica oil emulsion(all P<0.05).In the control group,cells grew well,there were a larger number of cells,with a normal growth state in shape.The number of cells in the cisplatin group and the Brucea Javanica oil emulsion group decreased in comparison with that of the control group,with a small number of debris visible.In the Brucea Javanica Ja-vanica oil emulsion and cisplatin group,the cell growth was significantly inhibited,the number of cells was the smallest,the shape of cells was irregular and the cell debris increased.The proportion of cells in the G0/G1 phase was as follows:Brucea Javanica oil emulsion + cisplatin group>cisplatin group,Brucea Javanica oil emulsion group>control group,the apoptosis rate was as follows:Brucea Javanica oil emulsion + cisplatin group>Brucea Javanica oil emulsion group,cisplat-in group>control group,and the intracellular ROS levels were as follows:Brucea Javanica oil emulsion + cisplatin group>Brucea Javanica oil emulsion group,cisplatin group>control group(all P<0.05).Conclusion Brucea Javani-ca oil emulsion can increase the sensitivity of lung cancer A549 cells to cisplatin chemotherapy,and its mechanism may be related to blocking tumor cells in G0/G1 phase,increasing the production of ROS,and promoting the apoptosis.
