非小细胞肺癌组织中组蛋白去乙酰化酶7、泛素特异性肽酶10的表达变化及其意义

目的 观察非小细胞肺癌(NSCILC)组织中组蛋白去乙酰化酶7(HDAC7)、泛素特异性肽酶10(USP10)的表达变化,探讨其意义。方法 98例NSCLC患者,均行肿瘤切除术,术中保留癌组织及癌旁组织。采用免疫组织化学法检测癌组织及癌旁组织HDAC7、USP10,采用Spearman相关性分析法分析NSCLC癌组织中HDAC7与USP10表达的相关性,分析HDAC7、USP10表达与NSCLC患者临床病理参数的关系,采用Kaplan-Meier生存曲线分析HDAC7、USP10表达与NSCLC患者预后的关系,采用COX多因素比例风险模型分析NSCLC患者预后的影响因素。结果 NSCLC组织、癌旁组织中HDAC7阳性率分别为65。31%(64/98)、6。12%(6/98)(χ2=74。756,P<0。05);NSCLC组织、癌旁组织中USP10阳性率分别为63。27%(62/98)、8。16%(8/98)(χ2=64。800,P<0。05)。肿瘤低分化程度、有淋巴结转移及TNM分期Ⅲ期NSCLC癌组织HDAC7,USP10表达阳性率高于高中分化程度、无淋巴结转移及TNM分期Ⅰ~Ⅱ期(P均<0。05)。NSCLC组织中HDAC7、USP10的表达呈正相关(r=0。714,P<0。05)。与表达阴性者比较,HDAC7、USP10表达阳性的患者3年累积生存率低(χ2=16。300、15。870,P均<0。05)。HDAC7表达阳性、USP10表达阳性、肿瘤低分化程度、有淋巴结转移、TNM分期Ⅲ期是NSCLC患者预后的独立危险因素。结论 NSCLC癌组织中HDAC7、USP10高表达,HDAC7、USP10可能协同促进NSCLC的发生发展,HDAC7、USP10高表达NSCLC患者的预后较差。
Expression changes of HDAC7 and USP10 in non-small-cell lung cancer tissues and their significance
Objective To investigate the expression changes of histone deacetylase 7(HDAC7)and ubiquitin-spe-cific peptidase 10(USP10)in non-small-cell lung cancer(NSCLC)tissues and their significance.Methods Ninety-eight NSCLC patients underwent tumor resection,with cancer tissues and adjacent tissues preserved during the opera-tion.Immunohistochemistry was used to detect the expression of HDAC7 and USP10 in tissues.Spearman correlation analysis was used to analyze the correlation between HDAC7 and USP10 expression in NSCLC cancer tissues,and to an-alyze the relationships between HDAC7 and USP10 expression and the clinicopathological parameters of NSCLC pa-tients.Kaplan-Meier survival curve was used to analyze the relationship between HDAC7 and USP10 expression and the prognosis of NSCLC patients,and the COX multivariate proportional hazard model was used to analyze the factors for the prognosis of NSCLC patients.Results The positive rates of HDAC7 in the NSCLC tissues and adjacent tissues were 65.31%(64/98)and 6.12%(6/98),respectively(χ2=74.756,P<0.05).The positive rates of USP10 in the NSCLC tissues and adjacent tissues were 63.27%(62/98)and 8.16%(8/98),respectively(χ2=64.800,P<0.05).The posi-tive expression rates of HDAC7 and USP10 in cancer tissues with stage III NSCLC,low tumor differentiation,and lymph node metastasis were higher than those with TNM stage Ⅰ-Ⅱ,high and medium differentiation,and no lymph node metastasis(all P<0.05).The expression of HDAC7 and USP10 in NSCLC cancer tissues showed a significant positive correlation(r=0.714,P<0.05).The 3-year cumulative survival rates of HDAC7 positive and USP10 positive patients were lower in comparison with those of HDAC7 negative and USP10 negative patients(χ2=16.300,15.870,respective-ly;all P<0.05).Positive HDAC7 expression,positive USP10 expression,poorly differentiated tumor,lymph node me-tastasis,and TNM stage III were independent risk factors for the prognosis of NSCLC patients.Conclusion HDAC7 and USP10 are highly expressed in NSCLC tissues,HDAC7 and USP10 may synergistically promote the development and progression of NSCLC,and the prognosis of NSCLC patients with high expression levels of HDAC7 and USP10 is poor.
