Association of HLA-DQA1*0501 and HLA-DQB1*02 gene polymorphisms with susceptibility to anti-Ro/SSA antibody-positive pSS
Objective To investigate the correlation between human leukocyte antigen(HLA)-DQA1*0501 and HLA-DQB1*02 gene polymorphisms and susceptibility to primary Sjögren's syndrome(pSS)in anti-Ro/SSA antibody-pos-itive individuals using database-related data.Methods The study screened and collected the data of HLA-DQA1*0501 and HLA-DQB1*02 gene polymorphisms in patients with pSS who were positive or negative for anti-Ro/SSA antibodies,as well as in healthy controls,using a computerized search of relevant databases.The associations between HLA-DQA1*0501 and HLA-DQB1*02 gene polymorphisms and the occurrence of pSS in anti-Ro/SSA antibody-positive patients was ana-lyzed using STATA 16.0(USA)statistical software.Results The study included five papers involving 420 patients with pSS,including 250 patients who tested positive for anti-Ro/SSA antibodies,120 patients who tested negative for anti-Ro/SSA antibodies,and 733 healthy controls.Of the pSS patients,159 and 246 cases tested positive for the HLA-DQA1*0501 and HLA-DQB1*02 genes,respectively.Among the healthy controls,196 and 282 cases tested positive for the HLA-DQA1*0501 and HLA-DQB1*02 genes,respectively.Among the pSS patients positive for anti-SSA antibodies,129 and 158 cases were positive for the HLA-DQA1*0501 and HLA-DQB1*02 genes,respectively.In the anti-SSA antibody-nega-tive pSS patients,30 and 46 cases were positive for the HLA-DQA1*0501 and HLA-DQB1*02 genes,respectively.The study found that HLA-DQA1*0501 and HLA-DQB1*02 gene polymorphisms were associated with susceptibility to pSS(I2= 62.99%and 40.75%,combined OR=2.60 and 2.43,95%CI=1.49-4.55 and 1.88-3.14,respectively;all P<0.05).The HLA-DQA1*0501 and HLA-DQB1*02 gene polymorphisms were also associated with susceptibility to anti-Ro/SSA an-tibody-positive pSS(I2=0.00%and 9.41%,combined OR=3.85 and 2.61,95%CI=1.81-8.21 and 1.52-4.48,respec-tively;all P<0.05).Conclusions HLA-DQA1*0501 and HLA-DQB1*02 gene polymorphisms are associated with sus-ceptibility to pSS in individuals who test positive for anti-Ro/SSA antibodies.Specifically,patients with anti-Ro/SSA anti-bodies and the HLA-DQA1*0501 and HLA-DQB1*02 genes are at higher risk of developing pSS.
