高脂饮食诱导肥胖小鼠不同脂肪组织炎症反应相关基因表达及其机制

目的 探讨高脂饮食诱导肥胖小鼠不同脂肪组织炎症反应相关基因表达及其机制。方法 将10只小鼠随机分为正常饮食对照组(ND组)、高脂饮食模型组(HFD组)各5只,分别喂以常规饲料、高脂饲料,饲喂10周。将小鼠颈部脱臼处死,取肩胛间棕色脂肪组织(BAT)、腹股沟白色脂肪组织(iWAT)、附睾白色脂肪组织(eWAT)。将各脂肪组织进行转录组测序(seq),并对测序结果进行质量评估以及主成分分析(PCA)。筛选三种脂肪组织中的差异表达基因,并对其进行基因本体论(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析;筛选表达上调最明显的前20个炎症反应相关基因。结果 HFD组较ND组体质量水平高(P<0。05)。两组脂肪组织seq质量评估结果显示错误率<0。1%,Q20>85%,Q30>80%。PCA显示两个主成分方差贡献率分别为51。81%、19。08%。iWAT中HFD组表达上调基因2 548个,eWAT中HFD组表达上调基因3 384个,BAT中HFD组表达上调基因1 630个;iWAT中HFD组表达下调基因2 082个,eWAT中HFD组表达下调基因2 780个,BAT中HFD组表达下调基因1 128个。在三种脂肪组织表达上调基因中,主要富集在炎症反应和免疫反应等过程;在三种脂肪组织全部表达下调基因中,主要富集在脂质代谢和小分子代谢等过程。HFD组富集通路主要为经典炎症信号通路NF-kappa B、PI3k-Akt、Toll-like receptor。结论 高脂饮食诱导肥胖小鼠的三种脂肪组织均存在炎症反应相关差异表达基因,而针对炎症反应会有不同的应答机制。
Expression of inflammatory response-related genes in different adipose tissues of mice with high-fat diet-induced obesity and its mechanism
Objective To explore the inflammatory response-related genes of different adipose tissues in mice with high-fat diet-induced obesity and its mechanism.Methods Ten mice were randomly divided into the normal diet control group(ND group)and high fat diet model group(HFD group),with 5 mice in each,which were fed with normal diet and high fat diet,respectively,for 10 weeks.The mice were killed by neck dislocation,and the interscapular brown adipose tissue(BAT),inguinal white adipose tissue(iWAT)and epididymal white adipose tissue(eWAT)were removed.The ad-ipose tissue was sequenced by transcriptome sequencing(seq),and the sequencing results were evaluated as well as prin-cipal component analysis(PCA).The differentially expressed genes in three kinds of adipose tissues were screened and an-alyzed by Gene Ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis,and the first 20 inflammatory related genes with the most up-regulated expression were screened.Results The level of body mass in the HFD group was significantly higher than that in the ND group.The re-sults of seq quality evaluation of adipose tissue in both groups showed that the error rate was<0.1%,with Q20>85%,and Q30>80%.PCA showed that the variance contribution rates of the two principal components were 51.81%and 19.08%,respectively.There were 2 548 up-regulated genes in iWAT of the HFD group,3 384 up-regulated genes in eWAT of the HFD group,1 630 up-regulated genes in BAT of the HFD group,2 082 down-regulated genes in iWAT of the HFD group,2 780 down-regulated genes in eWAT of the HFD group,and 1 128 down-regulated genes in BAT of the HFD group.Among the three kinds of up-regulated genes in the adipose tissues,they were mainly enriched in the processes of inflammation and immune response,while in all the down-regulated genes in three kinds of adipose tissues,they were mainly enriched in the processes of lipid metabolism and small molecule metabolism.In the HFD group,the main enrich-ment pathways were classical inflammatory signal pathways NF-kappa B,PI3k-Akt and Toll-like receptor.Conclusion There are differentially expressed genes related to inflammation in the three kinds of adipose tissues of obese mice induced by high-fat diet,and there are different response mechanisms for inflammation.
