Effects of intraperitoneal injection of resveratrol on mitochondrial biogenesis and apoptosis in embryonic spinal cord tissues of rats with neural tube defects
Objective To observe the effects of intraperitoneal injection of resveratrol(RSV)on mitochondrial bio-genesis regulatory factors and apoptosis related-proteins in embryonic spinal cord tissues of neural tube defects(NTDs)rat models.Methods Thirty female Wistar rats were randomly divided into the control group,teratogenic group,and RSV group.On the 10th day after conception,rats in the teratogenic and RSV groups were orally administered all-trans retinoic acid(atRA)to establish NTDs rat models.From the 8th to the 17th day after conception,rats in the RSV group were in-jected with RSV intraperitoneally once a day.On the 18th day after conception,embryos were removed by cesarean section in rats of each group,and spinal cord tissues were separated.Real-time fluorescence quantitative PCR was used to detect the mRNA levels of mitochondrial biogenesis biomarkers,including silent mating-type information regulation 2 homolog-1(SIRT1),transcription factor EB(TFEB),peroxisome proliferator-activated receptor gamma coactivator-1α(PGC-1α),and mitochondrial transcription factor A(TFAM)in rat embryonic spinal cord tissues.Western blotting was used to detect the protein levels of mitochondrial biogenesis biomarkers SIRT1,TFEB,PGC-1α,TFAM,and apoptosis-related proteins Bcl-2,Bax in rat embryonic spinal cord tissues.Results The relative expression levels of SIRT1 mRNA,TFEB mRNA,and TFAM mRNA in the teratogenic group were all lower than those in the control group(all P<0.05).The relative expres-sion levels of SIRT1 mRNA,Tfeb mRNA,PGC-1α mRNA,and TFAM mRNA in the RSV group were all higher than those in the teratogenic group(all P<0.05).The relative expression levels of SIRT1 protein,TFEB protein,PGC-1α protein,and TFAM protein in the rat embryonic spinal cord tissues of the teratogenic group were all lower than those in the control group(all P<0.05).The relative expression levels of SIRT1 protein,TFEB protein,PGC-1α protein,and TFAM protein in the rat embryonic spinal cord tissues of the RSV group were all higher than those in the teratogenic group(all P<0.05).Compared with the control group,the relative expression level of Bcl-2 protein in the rat embryonic spinal cord tissue of ter-atogenic group decreased,while the relative expression level of Bax protein increased(both P<0.05).Compared with the teratogenic group,the relative expression level of Bcl-2 protein in the rat embryonic spinal cord tissues of RSV group in-creased(P<0.05).Conclusion Intraperitoneal injection of RSV can increase the expression levels of mitochondrial bio-genesis regulatory factors and anti-apoptotic protein Bcl-2 in the embryonic spinal cord tissues of NTDs rat models.
