Relationships between miR-155-5p and SOCS1 levels and the degree of lesion and Th17/Treg imbalance in OLP patients
Objective To investigate the relationships between the levels of serum microRNA-155-5p(miR-155-5p)and suppressor of cytokine signaling1(SOCS1)and the degree of lesion and the imbalance of helper T cell 17(Th17)/reg-ulatory T cell(Treg)in oral lichen planus(OLP)patients.Methods Seventy-six OLP patients(observation group)were selected,including 28 non-erosive OLP patients and 48 erosive OLP patients(29 mild to moderate erosive OLP patients,19 severe erosive OLP patients),and meanwhile,50 healthy volunteers(control group)were randomly selected during the same period.Peripheral venous blood of all subjects was collected.Real-time fluorescence quantitative PCR was used to detect serum miR-155-5p and SOCS1 mRNA,and flow cytometry was used to detect the of Th17 ratio and Treg ratio,and to calculate the Th17/Treg.Pearson/Spearman correlation analysis was used to analyze the relationship between serum miR-155-5p level and SOCS1 mRNA level,and their relationships with the ratios of Th17,Treg and Th17/Treg in the peripheral blood.Results Compared with the control group,the serum miR-155-5p level,Th17 ratio and Th17/Treg in peripheral blood increased,while the serum SOCS1 mRNA level and Treg ratio in peripheral blood decreased in the observation group(all P<0.05).In patients with non-erosive,mile-to-moderate erosive and severe erosive OLP,serum miR-155-5p levels,Th17 ratio and Th17/Treg in peripheral blood gradually increased,while serum SOCS1 mRNA level and Treg ratio in peripheral blood gradually decreased(all P<0.05).Correlation analysis showed that the serum miR-155-5p level was negatively correlated with serum SOCS1 mRNA level in OLP patients(r=-0.809,P<0.01).Serum miR-155-5p level was positively correlated with Th17 ratio and Th17/Treg in peripheral blood(r/rs=0.819 and 0.820,both P<0.05),and was negatively correlated with Treg ratio(r=-0.735,P<0.05).Serum SOCS1 mRNA level was nega-tively correlated with Th17 ratio and Th17/Treg in peripheral blood(r/rs=-0.770 and-0.790,respectively;both P<0.05),and was positively correlated with Treg ratio(r=0.733,P<0.05).Conclusion The serum miR-155-5p level increased and serum SOCS1 mRNA level decreased in OLP patients,and the changes of both levels were closely related to the severity of disease and Th17/Treg imbalance.
