Vaginal flora diversity,changes in differential metabolic components and their related metabolic pathways in patients with cervical epithelial cell HPV-positive bacterial vaginitis
Objective To observe the vaginal flora diversity and metabolic component changes in cervical epitheli-al cell human papillomavirus(HPV)-positive bacterial vaginitis patients,and to explore potential biomarkers in HPV-positive BV patients.Methods Six patients with HPV-positive BV infection(observation group)and six healthy peo-ple(control group)were selected.Vaginal secretion samples were collected,and 16S rDNA gene sequencing technology was used to detect the vaginal flora of the two groups.Through the species composition,α diversity,and β diversity was used to analyze the vaginal flora diversity.The linear discriminant analysis was used to the effect size to screen the differ-ential microbiota between the two groups.Liquid chromatography-mass spectrometry technology was used to detect the vaginal secretions of two groups.The V16.0.2 SIMCA software was used to screen the differential metabolic components of the vaginal microbiota with variable information processing multiples of>1 and P<0.05 in the orthogonal partial least squares discriminant analysis,the pathway enrichment analysis of the differential metabolic components of the vaginal flo-ra in patients with HPV-positive BV infection in cervical epithelial cells was carried out through the Kyoto Encyclopedia of Genes and Genomes database,and the main metabolic pathways of P<0.05 were obtained.Results There were 201 operational taxonomic units(OTUs)in the observation group and the control group,with 526 and 293 unique OTUs,re-spectively.In the observation group and control group,the Shannon indexes were 3.93±1.11 and 1.65±0.85,Simpson indexes were 0.84±0.14 and 0.41±0.21,pielou_e indexes were 0.52±0.13 and 0.23±0.10(t=0.130,0.122,and 0.122,respectively;all P<0.05).The result of jaccard anosim test for the similarity of the two groups of vaginal flora was R=0.3726,P<0.05.Compared with the control group,the abundance of vaginal Fusobacterium increased at the phylum level,the abundance of Sneathia,Atopobium increased,and the abundance of Streptococcus decreased at the ge-nus level(all P<0.05);the differential species of the vaginal microbiota in the two groups were Fusobacteria,Fusobacte-riia,Fusobacteriales,Leptotrichia,Atopobium,Mageeibacillus,and Shuttleworthia.Compared with the control group,24 differential metabolic components of vaginal flora in the observation group were uracil,methylmalonic acid,nicotinic acid,etc.,and 6 differential metabolic pathways were pyrimidine metabolism,nicotinic acid and nicotinamide metabo-lism,etc.Conclusions The vaginal microbiota diversity of patients with cervical epithelial cell HPV-positive BV infec-tion increases,with mainly changes in Leptotrichia,Atopobium and Streptococcus.The differential metabolic compo-nents of the vaginal flora in HPV-positive BV patients include uracil,methylmalonic acid and nicotinic acid,etc;the dif-ferential metabolic pathways mainly include pyrimidine metabolism,nicotinic acid and nicotinamide metabolism path-way,etc.
