Ameliorative effect and mechanism of resveratrol on cardiac structure and function in mice with polycystic ovarian syndrome
Objective To investigate the ameliorative effect of resveratrol(Res)on the cardiac function and struc-ture of mice with polycystic ovarian syndrome(PCOS)and to explore its possible mechanism of action.Methods Thirty 3-week-old female SPF mice were divided into the model group,intervention group,and blank group.Mice in the model group and the intervention group were injected subcutaneously with dehydroepiandrosterone(6 mg/100 g)once a day for 21 consecutive days to construct the PCOS models,and the mice in the blank group were injected with 6 mg/100 g sesame oil subcutaneously(once a day,for 21 days).The mice in the intervention group were gavaged with 100 mg/kg Res once a day for 28 days,and the mice in the model group and the blank group were fed normally.Animal electrocardiographs and echocardiography were used to detect heart rate,QT interval and ejection fraction in each group.The mice in each group were sacrificed,the heart tissue was weighed and the aortic blood was retained,the hypertrophy of myocardial tissue was observed by HE staining,the fibrosis of myocardial tissue was observed by Masson staining,the M1 macrophage marker CD86 and M2 macrophage marker CD206 were detected by Western blotting,and the serum inflammatory cytokines inter-leukin 6(IL-6)and arginase 1(ARG-1)were detected by ELISA.Results Compared with the blank group,the model group had a lower heart rate,longer QT interval,lower ejection fraction and higher heart mass after the last gavage(all P<0.05).Compared with the model group,the intervention group had a higher heart rate,shorter QT interval,increased ejection fraction and lower heart mass after the last gavage(all P<0.05).Compared with the blank group,the cross-sec-tional area of heart cells in mice of the model group was larger and there were more fibrous tissues;compared with the mod-el group,the cross-sectional area of heart cells in the intervention group became smaller and the fibrous tissue decreased.Compared with the model group,the relative expression levels of CD206 protein in myocardial tissues were higher and the relative expression levels of CD86 protein were lower in the blank and intervention groups(all P<0.05).Compared with the blank group,the level of IL-6 in aortic serum of mice in the model group increased and the level of ARG-1 decreased(both P<0.05);compared with the model group,the level of IL-6 in aortic serum of mice in the intervention group de-creased and the level of ARG-1 increased(both P<0.05).Conclusions Intragastric administration of Res can improve the cardiac structure and function of PCOS mice,promote the expression of CD206 protein,inhibit the expression of CD86 protein in myocardial tissues,reduce the level of IL-6 in aortic serum,and increase the level of ARG-1 in aortic serum.The mechanism may be that Res promotes the M2 polarization of cardiac macrophages and alleviates the inflammatory re-sponse in cardiac tissues.
