Effect of curcumin on expression of matrix metalloproteinase and fibronectin in human retinal pigment epithelium
Objective To observe the effect of curcumin on the expression of matrix metalloproteinase(MMP)and fibronectin 1(FN1)in human retinal pigment epithelium(RPE).Methods Human RPE cells were divided into two groups:the curcumin group and control group;cells in the curcumin group were added with 0.5×10-5 mol/L curcumin,and cells in the control group were not treated with curcumin.After cells of the two groups were continuously incubated for 48 h,real-time fluorescence quantitative PCR was used to detect the matrix metalloproteinase(MMP)1,MMP2,MMP3,MMP9,and FN1 mRNAs,and Western blotting was used to detect the MMP1,MMP2,MMP3,MMP9,FN1 proteins in cells.Results The relative expression levels of MMP1,MMP2,MMP3,MMP9,and FN1 mRNAs in cells in the curcumin group were 0.41±0.14,5.020.22,1.090.21,0.470.28,and 2.500.21,and those in the control group were 5.100.38,6.430.15,0.270.19,1.820.12,and 6.830.18,respectively,with statistically significant differences be-tween these two groups(all P<0.05).The relative expression levels of MMP1,MMP2,MMP3,MMP9,and FN1 pro-teins were 6 155.33124.01,6 208.00157.33,8 126.33235.78,3 541.67 130.16,3 684.3390.22,respectively,in the curcumin group,versus 16 207.33210.19,8 053.3361.98,2 543.67122.20,9 477.33108.68,and 8 218.00239.86,respectively,in the control group,with statistically significant differences between these two groups(all P<0.05).Conclusion Curcumin up-regulates the expression of MMP1,MMP2,MMP9,FN1 and down-regulates the expression of MMP3 in human RPE cells.
