Therapeutic effect and mechanism of ethanol extract of Knoxia roxburghii on mice with bronchial asthma
Objective To observe the improvement effect of intragastric administration of ethanol extract of Knoxia roxburghii on respiratory function in bronchial asthma mice and to investigate its possible mechanism.Methods Thirty-two Balb/c female mice were adaptively fed for 1 week and randomly divided into the control group,model group,dexa-methasone group and the ethanol extract of Knoxia roxburghii group,with 8 mice in each group.OVA sensitizing solution[OVA 1 mg/mL and Al(OH)3 5 mg/mL]was intraperitoneally injected into mice to establish the asthma models in the model group,dexamethasone group and the ethanol extract of Knoxia roxburghii group;mice in the ethanol extract of Knox-ia roxburghii group and the dexamethasone group were given intragastric administration of 200 mg/kg of ethanol extract of Knoxia roxburghii and 1 mg/kg of dexamethasone,respectively;mice in the normal control group and the model group were both given the same volume of normal saline.On the second day of the last nebulization stimulation in each group,the average enhanced pause(Penh)values after 0,6.25,12.5,25,50,and 100 mg/mL methachcholine(Mch)were measured using a whole-body plethysmography system.ELISA was used to detect serum total immunoglobulin E(IgE),ov-albumin specific-IgE(OVA-IgE),and Th1 type inflammatory factor gamma interferon(IFN-γ),Treg inflammatory inter-leukin-10(IL-10),and Th2 type inflammatory factors(IL-4,IL-5,IL-9,IL-13)in bronchoalveolar lavage fluid(BALF).After execution,we taken the middle lobe tissues of the right lung and observed the pathological changes of the lung tissues after HE and PAS staining.Results Compared with the control group,the Penh values of the model group mice increased after 25,50,and 100 mg/mL Mch treatment(all P<0.05).Compared with the model group,the Penh val-ues of the dexamethasone group and the ethanol extract of Knoxia roxburghii group decreased after 50 and 100 mg/mL Mch treatment(all P<0.05).Compared with the control group,the serum IgE and OVA-IgE levels of the model group in-creased,while the average levels of IL-10 and IFN-γ in BALF decreased,and the levels of IL-4,IL-5,IL-9,and IL-13 in-creased(all P<0.05).Compared with the model group,the levels of serum IgE and OVA-IgE in the dexamethasone group and the ethanol extract of Knoxia roxburghii group decreased,while the levels of IL-10 and IFN-γ in BALF increased on av-erage,and the levels of IL-4,IL-5,IL-9,and IL-13 decreased.Compared with the control group,the lung tissue of the model group mice showed pathological changes such as airway inflammation infiltration,goblet cell proliferation,and high secretion of mucus;compared with the model group,the pathological changes in the lung tissues of mice in the dexametha-sone group and the ethanol extract of Knoxia roxburghii group were significantly reduced.Conclusion Intragastric ad-ministration of the ethanol extract of Knoxia roxburghii helps reduce airway hyperresponsiveness,inflammatory response,and mucus secretion in the lung tissues of asthmatic mice,and its mechanism may be related to restoring the immune bal-ance of Th1/Th2.
