Relationships between serum miR-142-3p and HMGB1 expression and condition and prognosis in patients with acute pancreatitis
Objective To investigate the relationships between the expression of serum microRNA-142-3p(miR-142-3p)and high-mobility group box 1(HMGB1)and the condition and prognosis of acute pancreatitis(AP)patients.Methods Totally 253 AP patients(AP group)and 100 physically examined healthy individuals(control group)were se-lected.AP patients were divided into the mild AP group(73 cases),moderately severe AP group(99 cases),and severe AP group(81 cases)according to the severity of the disease,and they were divided into the death group(25 cases)and the survival group(228 cases)according to the living condition.The serum miR-142-3p was detected by real-time quanti-tative fluorescence PCR and serum HMGB1 was detected by enzyme-linked immunosorbent assay at the time of admission in the AP group and at the time of physical examination in the control group,respectively.The binding sites of miR-142-3p and HMGB1 were predicted by TargetScan database,and Pearson correlation was used to analyze the correlation of se-rum miR-142-3p and HMGB1 expression in AP patients.We collected information about the prognosis of patients with AP.Multifactorial Logistic regression was used to analyze the effects of serum miR-142-3p and HMGB1 on the prognosis;receiver operating characteristic curve was used to analyze the predictive value of serum miR-142-3p and HMGB1 levels on death in AP patients.Results Compared with the control group,serum miR-142-3p expression was reduced and HMGB1 expression was elevated in the AP group(both P<0.05).The miR-142-3p and HMGB1 had a binding site,and their serum expression was negatively correlated in AP patients(r=-0.732,P<0.05).Serum miR-142-3p expression de-creased sequentially in the mild AP group,moderately severe AP group,and severe AP group,and HMGB1 expression in-creased in turn(all P<0.05).The independent risk factors for death in patients with AP were severe AP,prolonged ICU stay,and elevated HMGB1,and the independent protective factor was elevated miR-142-3p(all P<0.05).The area un-der the curve of serum miR-142-3p combined with HMGB1 in predicting death in AP patients was 0.900,which was great-er than that of either alone(both P<0.05).Conclusion The changes of serum miR-142-3p and HMGB1 levels are relat-ed to the condition and prognosis of AP patients,and the detection of serum miR-142-3p combined with HMGB1 is of high value in predicting the prognosis of AP patients.
