荠菜饮片的粉末显微特征及其薄层色谱鉴别

目的:为进一步完善提高荠菜饮片的质量标准,对其粉末显微特征和薄层色谱鉴别方法进行研究.方法:采用光学显微镜对荠菜饮片的粉末显微特征进行研究,并以亮氨酸和缬氨酸为对照品、荠菜为对照药材,进行薄层色谱鉴别.结果:显微鉴别观察的特征明显,其中非腺毛、气孔可作为荠菜的主要鉴别点.采用薄层色谱硅胶G板,稀乙醇为提取溶剂,超声处理 20 min,以正丁醇-冰乙酸-水(体积比 4︰1︰1)为展开剂,以茚三酮试液为显色剂,105℃加热后置日光下检视,所得薄层色谱图斑点清晰,分离度较好.结论:方法稳定性高、操作易行、重复性好,可为荠菜饮片的粉末显微特征及薄层色谱鉴别制定质量标准提供实验依据.
Study on Powder Microscopic Characteristics and Thin Layer Chromatography Identification of Capsella Bursa-pastoris(L.)Medic
Objective:In order to improve the quality standard of Capsella bursa-pastoris(L.)Medic.,the powder microscopic characteristics and thin layer chromatography(TLC)identification methods were studied.Methods:The powder microscopic characteristics of Capsella bursa-pastoris(L.)Medic.were studied by optical microscope.Leucine,valine and Capsella bursa-pastoris(L.)Medic.were used as the reference materials in TLC identification.Results:The characteristics of microscopic identification were obvious,among them non-glandular hair and stomata can be used as the main distinguishing points of Capsella bursa-pastoris(L.)Medic.The parameters were determined as follows:silica gel G thin-layer plate,dilute ethanol was used as the extraction solvent,ultrasound for 20 minutes,n-butanol-glacial acetic acid-water(4:1:1)as the development agent,ninhydrin as chromogenic agent,and heating at 105℃until the spots showed clear color in the daylight.The chmmatographic spots were clear,without intererence and easy to recognize.Conclusion:The method is stable,simple,feasible and reproducible.It can provide experimental basis for powder microscopic characteristics and TLC identification of Capsella bursa-pastoris(L.)Medic.
