化妆品中EGCG的含量测定及其稳定性研究

建立高效液相色谱(HPLC)法检测EGCG含量的方法.采用ZORBAX Extend-C18(250 mm×4.6 mm,5μm)色谱柱,以甲醇(A相)-0.05%三氟乙酸水溶液(B相)为流动相进行梯度洗脱,洗脱时间 35 min,柱温 35℃,流速 1.0 mL/min,进样量 10 μL,检测波长 280 nm.在此条件下,EGCG的线性范围为 10~1 000 mg/L,R2=0.999 6,精密度(n=6)RSD为 1.25%,重复性(n=6)RSD为 0.84%,稳定性RSD为0.61%.EGCG平均加样回收率为 92.27%,RSD为1.18%.该方法有利于化妆品不同剂型及原料中EGCG的质量控制.对添加有EGCG原料的不同剂型化妆品进行稳定性考察,结果发现EGCG直接以纯品添加,稳定性要优于EGCG磷脂复合物,但随着时间推移,EGCG含量下降较大;另外,温度对EGCG含量影响很大,低温放置更有利于含量稳定.为保证化妆品产品的功效,EGCG作为原料,更要做到足量添加.
Research on Determination and Stability of EGCG in Cosmetics
A method for the detection of EGCG content by high-performance liquid chromatography(HPLC)was established.ZORBAX Extend-C18(250 mm×4.6 mm,5 μm)Column was used for gradient elution with methanol(phase A)-0.05%trifluoroacetic acid aqueous solution(phase B)as mobile phase.The elution time is 35 min.The column temperature is 35℃.The flow rate is 1.0 mL/min.The injection volume is 10 μL,and the detection wavelength is 280 nm.Under these chromatographic conditions,the linearity range of EGCG is 10~1 000 mg/L,R2=0.999 6,precision(n=6)RSD was 1.25%,repeatability(n=6)RSD was 0.84%,and stability RSD was 0.61%.The average recovery rate of EGCG is 92.27%.The RSD is 1.18%.This method is conducive to the quality control of EGCG in different dosage forms and raw materials of cosmetics.The stability of cosmetics with different dosage forms of EGCG raw materials was investigated,and the results showed that the stability of EGCG was better than that of EGCG phospholipid complex,but the EGCG content decreased greatly with the passage of time.In addition,temperature has a great influence on the EGCG content,and low temperature storage is more conducive to the content stability.In order to ensure the efficacy of cosmetic products,EGCG should be added in sufficient quantities as raw materials.
