Effect of Overexpression of AtAUR1 on Cell Cycle Regulation in Root Tips under UV-B Stress
As the hole in the ozone layer in the North and South Poles continues to grow,the crisis caused by UV-B radiation to the ecosystems of the entire planet is growing.Plants are affected by their particular lifes-tyles and are more susceptible to the effects of enhanced UV-B radiation.As an abiotic stress signal,UV-B radi-ation can cause damage to the DNA of plant cells,which can significantly inhibit plant growth,and even lead to plant death in severe cases.As an important protein in the regulation of mitosis,Aurora kinase is involved in the localization and separation of chromosomes and cytokinesis during mitosis,and previous laboratory studies have shown that overexpression of AUR1 can alleviate the damage caused by UV-B radiation.In this study,Arabidop-sis AUR1 overexpressing lines containing the CYCB1;1-GUS tag was obtained by sexual hybridisation.The tissue expression pattern of CYCB1;1 in the obtained transgenic lines after UV-B treatment was analysed by GUS stai-ning and RT-qPCR,and the experimental results proved that overexpression of AUR1 could up-regulate the gene expression of CYCB1;1 in the root tip cells of the plants,and the staining results after UV-B radiation treatment further proved that AUR1 was involved in the UV-B radiation root tip.The staining results after UV-B radiation treatment further proved that AUR 1 was involved in the regulation of mitotic cycle in root tips after UV-B radia-tion,which provided a theoretical basis for revealing the effect of overexpression of AUR1 on the regulation of mi-totic cycle in root tips.
