Based on the NLRP3/NF-κB/COX-2 signal pathway to explore Tongjing capsule pairs effect of primary menstrual pain on rats
Objective:To investigate the effect of Tongjing pain capsule on the primary menstrual pain NLRP3/NF-κB/COX-2 signaling pathway in rats and its related mechanisms.Methods:60 adult healthy and clean grade SD fe-male rats were not pregnant or mated,were selected and all were in the estrus phase.The experimental animals were divided into 6 groups,with 10 animals assigned to each group using a random number table method.The study groups were as follows:normal control group,model group,ibuprofen capsule group,high-dose group of Tongjing capsule,medium dose group of Tongjing capsule,and low-dose group of Tongjing capsule.Except for the blank group,all other groups were established with a model of cold coagulation,blood stasis,and dysmenorrhea.The blank group was given distilled water(1 ml/100 mg)by gavage daily,the model group was given distilled water(1 ml/100 g)by gavage,the low,medium,and high doses of Tongjing capsule were given corresponding doses by gavage,and the ibuprofen capsule group was given ibuprofen capsules(0.06 g/kg)by gavage continuously for 7 days.Observe the number of twisting reactions in each group of rats.Observation of uterine pathology by hematoxylin eosin(HE)stai-ning.Enzyme linked immunosorbent assay(ELISA)was used to detect the levels of prostaglandin F2α(PGF2α)and prostaglandin F2(PGF2).Western blot was used to detect the expression of NLRP3,NF-κB,and COX-2 proteins in rat uterine tissue;Real time fluorescence quantitative PCR(RT-PCR)was used to detect the mRNA expression of NLRP3,NF-κB,and COX-2 in rat uterine tissue.Results:Compared with the control group,the rats in the model group showed an increase in the number of twists,PGF2α,and OT levels(P<0.001),a decrease in PGE2 levels(P<0.001),mild dilation of uterine glands,infiltration of granulocytes and lymphocytes,and a significant upregula-tion of COX-2,NF-κB,and NLRP3 protein relative expression in uterine tissue(P<0.001),as well as a significant upregulation of COX-2,NF-κB and NLRP3 mRNA levels(P<0.001).Compared with the model group,the rats in each treatment group showed a significant decrease in the number of twists,PGF2α,and OT levels(P<0.05,P<0.01,or P<0.001),a significant increase in PGE2 levels(P<0.05,P<0.01,or P<0.001),clear uterine structure,intact endometrium,no obvious glandular hypertrophy or uterine myometrial hyperplasia,reduced inflammatory in-filtration,downregulation of COX-2,NF-κB and NLRP3 protein relative expression(P<0.05,P<0.01,or P<0.001),and down regulation of COX-2,NF-κB and NLRP3 mRNA levels(P<0.05,P<0.01,or P<0.001).Conclusion:Tongjing capsule can improve uterine inflammatory cell infiltration and act as analgesics by regulating the inflammatory response of menstrual pain through NLRP3/NF-κB/COX-2 signaling channels.
