聚己内酯/壳聚糖载药敷料的制备及凝血性能

伤口愈合是一个复杂、动态且缓慢的过程,为了加快这一进程,利用静电喷雾法制备了负载凝血酶的聚己内酯/壳聚糖碗状微粒.对碗状微粒的微观形貌、化学结构、凝血性能、血液相容性及细胞毒性进行了研究.结果表明:凝血酶@聚己内酯/壳聚糖碗状微粒具有良好的血液相容性和生物相容性,其溶血率低于2%,细胞存活率皆在95%以上;凝血酶极大地增强了聚己内酯/壳聚糖碗状微粒的凝血性能,凝血酶@聚己内酯/壳聚糖碗状微粒的活化部分凝血活酶时间和凝血酶时间仅为21.4 s和13 s;凝血酶@聚己内酯/壳聚糖碗状微粒涂层纱布的凝血指数远低于同时间普通纱布的凝血指数.
Preparation of polycaprolactone/chitosan drug-loaded dressing and its coagulation properties
Wound healing is a complex,dynamic and slow process.In order to accelerate this process,polycaprolactone/chitosan bowl-shaped particles loaded with thrombin are prepared by electrostatic spraying.The microstructure,chemical structure,coagulation properties,blood compatibility and cytotoxicity of the bowl-shaped particles are investigated.The results show that the thrombin@polycaprolactone/chitosan bowl-shaped particles have good blood compatibility and biocompatibility.The haemolysis rate is less than 2%and the cell survival rate is more than 95%.Thrombin can significantly enhance the coagulation properties of polycaprolactone/chitosan beads.The activated partial thromboplastin time and thrombin time of thrombin@polycaprolactone/chitosan bowl-shaped particles are only 21.4 s and 13 s,respectively;the coagulation index of thrombin@polycaprolactone/chitosan bowl-shaped microparticle-coated gauze is much lower than that of ordinary gauze at the same time.
